Objective:

The Center for Children’s Environmental Health Research at the University of California, Berkeley, is a fully coordinated, interdisciplinary research program that addresses the unique environmental health needs of children living in a primarily Latino, farmworker community. Over the past 12 years, our research has focused on pesticides: children’s routes and levels of exposure, the resulting health consequences, genetic susceptibility and mechanisms of effect, and community-based strategies to reduce exposure. We have developed strong community partnerships with extensive outreach to farmworkers, growers, service providers, and policy makers. We also have leveraged Center resources to obtain additional grants, including resources to examine exposure to polybrominated diphenyl ethers (PBDEs) in this population.

The keystone of our Center has been the CHAMACOS (Center for the Health Assessment of Mothers and Children Of Salinas) study, a longitudinal cohort of primarily low-income, Mexican immigrant farmworker women and their children living in the agricultural Salinas Valley, California. Women were enrolled during pregnancy and their children have been followed through 9 years of age, providing a unique opportunity to examine prospectively the influence of prenatal and early childhood exposures on children’s health. In this, our third phase of the Center, we are following the CHAMACOS boys from age 9 to age 13 years and we are working to double the size of the cohort to 300 boys (girls are being followed under a separate grant). We have expanded our focus to a “California mix” of chemicals: dichlorodiphenyltrichloroethane (DDT), to which the children were exposed in utero due to their mothers’ exposure in Mexico; manganese (Mn), which is a key component of the widely used agricultural fungicides maneb and mancozeb; and PBDE flame retardants, to which exposure is very high in these children due to California’s strict flammability standards. The Center is comprised of several research projects and cores.

Progress Summary:

This report does not cover a full year. It covers the period 8/1/2010-5/31/2011.

Project A: “The CHAMACOS Cohort Project: Pesticides and PBDE on Neurobehavior & Puberty” (R834513C001)

Project A directly addresses worldwide concerns that changes in onset of sexual maturation may be related to endocrine disruptors in the environment and fills a large data gap on boys. It also addresses concerns that exposure to DDT/E, PBDEs and Mn may compromise neurodevelopment.

Exposure to these chemicals is widespread. PBDEs have been detected in 97% of the U.S. population. Exposure to background levels of DDE also is ubiquitous in the United States and exposure to DDT can be very high in developing countries where DDT is increasingly used for malaria control. In the United States, exposure to manganese (Mn) occurs from industrial sources, but little research has been conducted on agricultural sources, even though approximately 9 million pounds of the manganese-containing fungicides mancozeb and maneb are applied annually in the United States. These exposures may be particularly relevant in agricultural communities in California, which have the potential for high exposure to all three compounds.

Animal studies suggest that PBDEs, DDT/E, and Mn may impact both neurodevelopment and pubertal development, but there have only been a limited number of human studies. This study will fill important data gaps in our knowledge of the health effects of these widespread exposures.

Aims: The specific aims of Project A have not changed this year.

In Project A, we are examining the association of DDT, Mn, and PBDEs with neurodevelopment and onset of puberty in boys in the CHAMACOS cohort.

  1. To maintain and expand the CHAMACOS cohort as children begin the critical transition to puberty, assessing neurodevelopment and pubertal development in 300 boys from 9 to 13 years of age.
  2. To determine whether prenatal and childhood exposure to DDT/E, PBDEs, and Mn are associated with neurobehavioral functioning at age 9, 10½, and 12 years.
  3. To determine whether prenatal and childhood exposure to DDT/E, PBDEs, and Mn are associated with timing of pubertal development in boys between age 9 and 13 years.
  4. To determine whether prenatal and childhood exposure to DDT/E, PBDEs, and Mn are associated with hormone levels in boys at age 12.

Project B: “Exposure Project: Mn, DDT/E and PBDE Exposure to Farmworker Children” (R834513C002)

In Project B, measurements of Mn in deciduous tooth cross sections will be validated as a new biomarker of Mn exposure in children, substantially contributing to the field of metal exposure and environmental epidemiology. Agricultural use of Mn-containing fungicides is a potential source of Mn in California and nationally. Determining whether these pesticides contribute to human exposures and health effects is an important public health issue. Studies of PBDE exposure will inform researchers about the impact of California flammability laws.

In Project B, we will examine novel methods of examining prenatal exposure to Mn, PBDE, and DDT/E compounds using shed deciduous teeth and GIS methods with remote sensing.

  1. To measure Mn, PBDEs, and DDT/E in shed deciduous teeth. (Population: CHAMACOS children participating since birth.)
  2. To determine the relationship between Mn, PBDE, and DDT/E levels in teeth with other relevant biological measures.
  3. To quantify the contribution of agricultural pesticide use to Mn levels in house dust, teeth and child blood.
  4. To identify population correlates of PBDEs and DDT/E exposure in 9-year-old boys.

Project C: “Epigenetics Project: Epigenetic Effects of DDT/E and PBDEs on Puberty” (R834513C003)

In Project C, we are investigating the effects of exposure on the epigenome and its relationship with pubertal onset. This year, we have continued to assess global methylation in DNA isolated from banked clot specimens of CHAMACOS children using three different assay methods: pyrosequencing (Alu and LINE-1 repeats) and Illumina Infinium. Preliminary analyses suggest that global methylation patterns are likely fluctuating throughout early childhood. If verified through expanded analysis, the opposite directions of the change in global DNA methylation by LINE-1 and Illumina 27k assays may highlight the importance of using site specific methylation data. Furthermore, we have found that global DNA methylation may be modified by prenatal exposure to PBDEs, providing a potential mechanism by which PBDEs and possibly other toxicants affect health.

Aims: The specific aims of Project C have not changed this year.

  1. To analyze global DNA methylation in newborn children by three different assays.
  2. To determine ontogenetic changes in global DNA methylation in blood of children between birth and 12 years.
  3. To investigate the relationship of in utero and 9-year-old blood concentrations of DDT/E and PBDEs with global DNA methylation.
  4. To determine whether global methylation is associated with onset of puberty and hormonal changes.
  5. To examine site-specific methylation in relation to age, sex, exposure to DDT/E and PBDEs and puberty onset.

Community Outreach and Translation Core (COTC) (R834513C004)

COTC activities are vital to our Community-University partnership. The COTC permits Center scientists and community partners to communicate study findings in a culturally appropriate manner, to raise awareness of children’s environmental health within and beyond the Salinas Valley, and to mobilize groups toward actions that will improve the health of low-income Latino residents in Monterey County. The specific aims for the COTC remain unchanged.

  1. To disseminate Center research findings to study participants, the Salinas Valley community, and other stakeholders.
  2. To increase awareness about children’s environmental health among low-income Latino communities, clinicians, and service providers through widespread dissemination of innovative outreach and educational programs.
  3. To build the capacity of Salinas Valley youth to promote healthy environmental policies in their community.
  4. To educate policy makers at the local, state, and national levels about Center research findings and children’s environmental health priorities.

The Biorepository Core: In the past 12 years of Center activity, more than 100,000 environmental and biological specimens have been collected. In the Biorepository Core, our role is to assure that all such samples are collected, managed, and stored so as to maximize their future utility, as well as coordination of sample transfer to analytical laboratories for analysis. We have developed standard operating procedures (SOPs) with quality controls for each specimen type, and we developed comprehensive systems to track, process, and store samples. Each of the Center’s research projects utilizes the Biorepository Core’s stored biological or environmental samples for key exposure or health endpoint data. The Biorepository Core continues to provide critical support to the research projects in sample collection, processing, analysis, storage, tracking, and transport, and in use of banked specimens.

The Administrative Core: This core functions as a centralized facility that coordinates overall Center administration, financial and logistical operations, and allows investigators to concentrate on research. Moreover, this core provides key support necessary to facilitate communications between the Center offices in Berkeley and Salinas and among community partners, project and core directors, collaborators, internal and external advisory boards, NIEHS, EPA, and other participating institutions.

Future Activities:

Project A (R834513C001):

In year 3, we will complete recruitment of new CHAM2 participants, continue data collection at 10½ years, and begin the 12-year visit (in February 2012). Interim visits at 9¾ and 11¼ to assess pubertal development will be ongoing.

Project B (R834513C002):

In the next year, measurements of Mn in teeth, and for a subset, maternal and child blood and urine and breastmilk, will be completed. We will begin analyses examining the interrelationships of Mn in teeth and other biological samples as well as completing the analyses of predictors of Mn in house dust. We also will complete analysis of Mn and PBDEs in blood from CHAMACOS 9-year-old children.

Project C (R834513C003):

Next year, we will expand pyrosequencing and Infinium analyses to additional samples from the same children at other ages (2, 5, 9 years old), and confirm our preliminary finds for Alu, LINE-1 and Illumina for a larger sample size. Additionally, as CHAMACOS puberty data become available at the end of 2011, we will analyze how global and site-specific methylation changes through pubertal development.

COTC (R834513C004):

  • Dissemination of Center research findings: We plan to hold our annual community forum, continue targeted outreach to community groups, and respond to invitations for presentations. We also will continue our dissemination to the scientific community.
  • Education on children’s environmental health: Over the next year, we plan to set up the Prenatal Environmental Health Kiosk for use in additional health clinics and social service agencies. Environmental education to childcare providers and community groups will continue.
  • Youth Community Council (YCC): Over the summer, the YCC will work on a Photovoice Project where they will use cameras to record the positive and negative health impacts of their local environment. After the project is complete, they will present their work to a community audience and their art will be posted in a public space.
  • Policy Makers: We will continue to present our research to policy makers as requested. The Tri-County Association of Latino Elected Officials, a regional group, has invited us to describe the CHAMACOS studies and findings.
