Objective:
The central hypothesis of the Southern California Particle Center and Supersite (SCPCS) is that organic constituents associated with particulate matter (PM)—including quinones, other organic compounds (polycyclic aromatic hydrocarbons [PAHs], nitro-PAHs, and aldehydes/ketones), and metals—are capable of generating reactive oxygen species and acting as electrophilic agents. They have a central role in allergic airway disease such as asthma and cardiovascular effects through their ability to generate oxidative stress, inflammation, and immunomodulating effects in the lungs and airways.
Progress Summary:
This progress report addresses the activities of the Biostatistical Core. Dr. Daniel Stram continues to provide statistical consultation with projects supported by the SCPCS. Dr. Stram has been providing direct assistance to SCPCS Principal Investigators (PIs) in two main areas: analyses of human exposure studies conducted at Rancho Los Amigos (Dr. H. Gong, PI) and analysis of the data from Dr. Kleinman's freeway exposure study. During his sabbatical leave (June-December 2002), Dr. Brian Langholz served in his stead and mainly focused on the human exposure studies at Los Amigos.
Consultation on SCPCS Projects. Dr. Stram has become closely involved in the analysis and presentation of data from the human exposure studies at Rancho Los Amigos. He has rewritten critical portions of the Rancho Los Amigos analysis code, updating it from BMDP to a much better supported SAS procedure for repeated measures, SAS PROC MIXED, while verifying the reproducibility and appropriateness of the earlier analyses. He has updated the general approach to the graphical display of the data from exposure experiments taking place at Rancho so that plots generated more accurately represent the statistical findings of the repeated measures analyses. This new code now has been extensively used by himself and Dr. Langholz to analyze data from several of the human exposure studies.
Since returning from sabbatical, Dr. Stram has begun working closely with Dr. Kleinman on a cohesive analysis of preliminary data on the immunologic responses of mice to concentrated particles for the first several sites and years of exposure.
A manuscript presently is being written on the results of an exposure study of 40 volunteers at Los Amigos, including normal and chronic obstructive pulmonary disease patients, relating responses of interest to treatment conditions (exposure to particles or filtered air) and to ambient exposures in periods just before the day of the experiment. Dr. Gong has presented results from this analysis. Dr. Kleinman also has presented initial results of the freeway studies.
Future Activities:
Dr. Stram will continue to work closely with SCPCS investigators, especially on the development of manuscripts from the Los Amigos human and freeway mouse exposure studies. He will review all data management plans as well as analytic approaches of the SCPCS investigations.
