Objective:
The overall objective of the Southern California Particle Center (SCPC) is to bring together outstanding scientists to conduct high priority research to elucidate the underlying basis for health effects associated with exposure to ambient particulate matter (PM). The overall goal of the SCPC is to establish linkages between PM source emissions, underlying mechanisms of toxicity and resulting health effects. Fine and ultrafine particles derived from mobile sources continue to serve as foci for SCPC interdisciplinary studies that include exposure, toxicology, chemical characterization and epidemiology. Health effects research centers around identifying mechanisms of PM-induced cardiovascular effects and allergic airways disease and the exposure conditions and components that are associated with observed chemical and toxicological mechanisms.
The research of the Center is divided into five inter-related project areas. The primary objective of Project 1 is to examine the relationships between PM sources, exposure, and toxicity within the constraints of the urban atmosphere. This project is an integral part of Projects 2, 3 and 4, by serving as the field operations to collect PM samples for toxicity testing and for providing elevated levels of ambient PM for animal exposure models described in these projects. Our major themes in Project 1 are:
- Physical and chemical properties of PM emitted from different PM sources.
- To determine their emission rates of PM species vs size
- To evaluate how exposure to PM from these sources vary with respect to location, season, and particle size,
- In conjunction with Projects 2, 3 and 4 to assess their relative toxicity by providing in vitro PM samples to the PIs of these projects
- Determine the characteristics of the volatile and non-volatile particle components of these sources. Also provide in vitro samples to Projects 2- 3
- Measure exposure gradients and intra-community variability of PM from complex, unstudied sources such as airports and port activities.
- To assess the contributions of these outdoor sources to indoor exposure in support of Project 4.
Progress Summary:
In year 5 Project 2 continued our effort to answer the question whether inhalation of ambient UFP could boost the secondary immune response in previously sensitized animals during an allergen response. In collaboration with Project 1, we completed our “real-life” inhalation exposure studies in our mobile animal research laboratory located near the 110 freeway in downtown Los Angeles. Our data show that inhalation exposure to “real-life” UFP was sufficient to boost the secondary immune response during experimental allergen (OVA) challenge in prior sensitized animals, resulting in a profound allergic inflammation in alveolar duct and alveolar parenchyma of the lung, which is accompanied by a Th17 immune response and the induction local oxidative stress (Li, et al., Am J Physiol Lung Cell Mol Physiol, 2010). A proteome profiling of the bronchoalveolar lavage (BAL) and lung samples from this study is currently in progress. In collaboration with Dr. Joseph Loo from the Keck Proteomics Facility at UCLA, we have conducted proteomic profiling in year 5 in order to identify new proteins in the BAL that may be useful biomarkers for allergic inflammation related to air pollutants. Using our previously published intranasal sensitization mouse model, we have demonstrated that the adjuvant effect of UFP was closely correlated to a significant change in the proteome profile in BALF. The findings from this work have been published (Kang, et al., Proteomics, 2010).
We also have made progress in studying the role of dendritic cells (DC) in executing the adjuvant effects of DEP. We could demonstrate that DEP was capable of enhancing DC function which, upon adoptive transfer, could result in an enhanced allergic sensitization to OVA in the recipient mice, leading to an enhanced eosinophilic inflammation in the lung and increased OVA-IgG1 production in the plasma. In summary, the progress made by Project 2 demonstrates that ambient UFP could effectively promote allergic airway inflammation via the induction of oxidative stress, which may be reflected by altered proteome profiles of the lung.
Project 3 proceeded with the following hypothesis:
- PM contains pro-oxidative chemicals
- Organic chemicals and metals located on the PM matrix are responsible for toxicity
- PM generates ROS and electrophilic chemistry◊ oxidative stress including impact on cell signaling pathways
- Oxidative stress ◊Pro-inflammatory effects
Prooxidants and electrophiles, the reactive chemical species proposed to affect cellular processes have been found in both particles and vapors of diesel exhaust and ambient air. Samples collected from diesel vehicles operating in a dynamometer system contained mostly organic prooxidants that can be removed by heating to temperatures of 100 to 250° C. The particle phase of ambient air contains most of the prooxidants, both organic and metallic. The volatile organic components, collected on XAD resin traps, contained most of the electrophiles of ambient air. Thus, exposure to ambient air involves metals and organic prooxidants that can deposit in the lungs, allowing direct interaction between the particle and the cell. The particle can serve as a “carrier” allowing the penetration of the cell of particle bound metals and organic compounds by phagocytotic processes. Once inside the cell, the adsorbed chemicals could dissociate into the cell milieu and initiate prooxidant actions. The reactive organic vapor phase components, mostly electrophiles, could enter cells by passive diffusion and form covalent bonds with nucleophilic centers in proteins, initiating a different set of cellular responses.
Cellular responses to 1,2-naphthoquinone (1,2-NQ) have led to the identification of two key proteins, PTP 1B and Keap 1, regulatory proteins for the EGFR and ARE, respectively as targets. Activation of these targets lead to cell proliferation and inflammation on the one hand, and protective proteins on the other. In this interaction, 1,2-NQ is acting as an electrophile, since the oxidative stress levels were low. Analogous experiments with the vapor phase components of ambient air samples have shown similar effects. Thus, the net effect of exposure to both particles and vapors will reflect the actions of prooxidant metals and organic compounds, as well as electrophiles.
Project 4 investigators prospectively followed a cohort panel of elderly people with coronary artery disease living in retirement communities of the Los Angeles air basin. They found positive associations of circulating biomarkers of inflammation and platelet activation in plasma with both quasi-ultrafine particle mass and markers of exposures linked to fossil fuel combustion (including PM2.5 carbonaceous aerosols), but not with secondary (photochemically-related) organic carbon or ozone. In a majority subset of this panel, they also found that erythrocyte antioxidant enzymes decreased with exposure to the same air pollutants. Since the erythrocyte antioxidant enzymes were also inversely associated with circulating biomarkers of inflammation, it is possible that enzyme inactivation may play a role in the pro-inflammatory effects of air pollutants. In vitro work from Projects 3 and 4 suggest that particle electrophilic potential can lead to the irreversible inactivation of protein function, including antioxidant enzymes. A follow-up analysis in Project 4 found that the PAH fraction of quasi-ultrafine particles explained associations of biomarkers of inflammation with quasi-ultrafine mass. PAH was linked to combustion emissions from traffic using chemical mass balance model that included hopanes (vehicle emissions tracer). Interestingly, markers of secondary organic aerosols in quasi-ultrafine particles (water-soluble carbon and n-alkanoic acids) were not associated with circulating biomarkers of inflammation, but were associated with exhaled nitric oxide, which is a biomarker of airway inflammation. This suggests that different particle components affect different target organs likely due to differing toxicokinetics. In the same panel, Project 4 investigators found significant and large increases in systolic and diastolic blood pressure in relation to PM2.5 mass, black carbon, and primary organic carbon exposures. Blood pressure was more weakly associated with secondary organic carbon, and was not associated with ozone. In a parallel analysis of ambulatory electrocardiograph data from 38 of the subjects in the panel, Project 4 investigators found significant associations between the risk of ST segment depression ≥1.0 mm with exposure to quasi-ultrafine particles and markers of primary products of fossil fuel combustion. ST segment depression was not associated with PM2.5 secondary organic carbon or ozone. These results suggest that primary products of fossil fuel combustion lead to an increased risk of myocardial ischemia. Overall, these significant findings for quasi-ultrafine particles and markers of traffic-related particles support the hypothesis that redox-active and other particle components from the combustion of fossil fuel directly affect cardiovascular target sites resulting in systemic inflammation and platelet activation. As a possible result of this and other mechanisms, these same exposures are significantly associated with adverse cardiovascular responses including increased blood pressure and electrocardiographic evidence of cardiac ischemia.
As part of our ongoing effort to determine the relative contributions of vapor and particle components of ambient air samples to the health effects of urban air pollution we undertook a sampling campaign to obtain simultaneous large samples (250-500 m3) of particles and vapor phase components from the same volume of ambient air. Samples were taken at three different locations having a different mix of fresh, aged, and photochemically produced contaminants: a freeway adjacent site, an urban site and a receptor site. Both phases were used for the full slate of bioassays and detailed chemical analysis. Particle phase showed greater redox activity than vapor phase components, but the vapor phase showed higher electrophilic content then the particle phase. Quinone and quinone-like compounds are important contributors to vapor phase toxicity.
A related project assessing the cardiovascular changes associated with short-term exposure to freeway air among persons over 60 found a significant increase in atrial arrhythmias during and following a two-hour exposure to freeway air compared to similar exposure to filtered freeway air. Also blood samples from the subjects showed significant increases in two biomarkers following exposure to unfiltered air compared to filtered air: N-terminal proB-type natriuretic peptide (NT pro-BNP) increased 38% and vascular endothelial growth factor (VEGF) increased 30%. These effects were strongest for ultrafine particles.
