Objective:
The principal concern of the UC Davis Center for Children’s Environmental Health (CCEH) is to identify and understand environmental, immunologic, and genetic risk factors contributing to the incidence and severity of childhood autism. Autism is a heterogeneous neurodevelopmental disorder defined by core deficits in social reciprocity, communication, and restrictive/repetitive patterns of interest and behavior. Generally accepted estimates of prevalence range from 1:750 for the narrowest diagnostic criteria, to 1:166 for autism spectrum disorder (ASD). The California Department of Developmental Services records on average approximately 700-750 autism diagnoses each quarter. Although autism may be one of the most heritable complex disorders, the defective genes conferring autism risk do not segregate in a simple Mendelian manner. The prevalence of autism is approximately four times higher in males than in females. Results from over 10 genome-wide autism screens indicate that potential susceptibility genes are spread across the entire genome. Estimates of the number of genes involved in autism range from 3-10 to 15 and up to 100. However, no single locus alone appears to be sufficient to cause the full clinical phenotype. Evaluation of a broader autistic phenotype that included communication and social disorders increased concordance from 60% to 92% in monozygotic twins and from 0% to 10% in dizygotic pairs. These results indicate that interactions among multiple genes are likely to contribute to autism, and that epigenetic factors and exposure to environmental modifiers may contribute significantly to variable expression of autism, and autism-related traits.
The mission of the CCEH is to promote daily interactions among a multidisciplinary team of scientists whose main research interest is to understand the complex web of etiologic factors that contribute to autism. The shared philosophy among Center participants is that a better understanding of the immunological and neurobiological mechanisms associated with this neurodevelopmental disorder can not only lead to a better understanding of the mechanisms that influence it but can also accelerate the discovery of effective intervention strategies.
Progress Summary:
Project 1 – Environmental Epidemiology
Description: Three epidemiological studies are being conducted: 1) The CHARGE (Childhood Autism Risks from Genetics and the Environment) Study, which was launched in the first 5-year period of the CCEH, continues in its second five-year funding cycle as a separately funded R01. By 2011, the study will include 1600 children and their families, the largest and most comprehensive study of environmental factors in autism. 2) The MARBLES (Markers of Autism Risk in Babies – Learning Early Signs) Study, a longitudinal cohort study of pregnancies of high-risk children was launched, after a pilot phase. The pilot phase began recruitment in July 2006, supported by funding from the M.I.N.D. Institute. 3) The CHARGE-BACK Study has recruited children and their immediate family members who participated in the CHARGE study in the first five years. The study is investigating whether immune function differences between study subject groups found in the first project period will be maintained over time. In addition, studies in the first project period were aimed at a broad analysis of immune function while the present study seeks to address the mechanisms responsible for the immune function anomalies.
Key findings:
Questionnaire results indicate that several food items and household furnishings treated with brominated flame retardants are associated with higher PBDEs. We published results showing that PBDE concentrations are related to food consumption, demonstrating that household furnishings are not the only major source in young children. Given the role of food, we hypothesize that the use of current plasma is not an adequate surrogate for prenatal exposures of the fetus, and hence more work is needed with better surrogates or actual measurements, as in the MARBLES study (Key Words: Persistent Organic Pollutants)
Our research on folic acid showed that mean folic acid intake was greater for mothers of typically developing children than for mothers of autistic children, especially in the month before and the first month of pregnancy. This difference was primarily due to a greater proportion of mothers of typically developing children consuming prenatal vitamins before and during the first month of pregnancy (adjusted for maternal). Women reporting the highest average intake of folic acid (800+ mcg) before and during the first month of pregnancy had the lowest risk of having children diagnosed with autism. Data from plasma specimens on close to 100 CHARGE study children showed no significant differences between cases and controls, but did show a correlation with several immune markers, including lower IgG, lower IgM and lower TGF-beta; all three of these immune outcomes were associated with case status in a larger sample of CHARGE children. Analysis of mercury levels indicated no differences between affected and unaffected children, after controlling for intake from fish and other sources. (Key Word: Mercury)
We also published results from two analyses pertaining to statewide autism incidence (one addressing parental age; the other, spatial clustering). (Key Word: Autism Onset, Regression)
Genotyping of some vitamin D genes has been completed and we will begin analyses of vitamin D in the Environmental Exposure Questionnaire soon, along with measuring vitamin D in plasma samples and newborn blood spots. (Key Words: Autism Nutritional Risk Factors)
Also, we collaborated with Dr. Xin Zhang, who conducted a case-control study of prenatal and perinatal risk factors for autism in Tianjin, China, and published those findings. (Key Words: Persistent Organic Pollutants)
In conjunction with the CCEH Genetics/genomics core, we published results on suggesting that the relationship of gene expression to mercury and lead differs by case status. Analyses of metals do not show any differences with regard to regression vs. early onset, language delay, nor cognitive or adaptive function. (Key Word: Mercury)
Project 2 – Immunological Susceptibility of Autism
Description – The goal of this project is to examine the effects of environmental toxicants on immune function in children with autism. In addition, we are currently working on the identification of the autoantigens in both the maternal population and their offspring. We will bring back our subjects for a longitudinal analysis of their immune function parameters studied previously.
Key Findings:
Effects of in vitro BDE 49 exposure: Our studies indicate that BDE49 has greater effects on immune function, particularly in the autistic population, at much lower concentrations than BDE47. (Key Words: Persistent Organic Pollutants)
Maternal Autoantibodies: The presence of these antibodies in the plasma of some mothers of children with autism, as well as the differential findings between mothers of children with early onset and regressive autism may suggest an association between the transfer of autoantibodies during early neurodevelopment and the risk of developing autism in some children. We have now identified several proteins recognized by maternal antibodies and are in the final stages of confirmation of these antigens. In addition, we are testing our plasma bank for reactivity to the individual antigens for all maternal samples. (Key Words: Immune Impairments)
Cytokine production: We published our findings describing plasma cytokine profile of subjects with autism and typically developing controls. Increasing cytokine levels were associated with more impaired social interactions and communication as well as more aberrant behaviors. It was noted that the increased cytokine levels were predominantly in autistic children who had evidence of regression. These findings suggest that ongoing inflammatory responses may be linked to disturbances in behavior and require confirmation in a larger study. Characterization of immunological parameters in autism has important implications for diagnosis, the designing and monitoring of therapeutic treatments of autism, and may help to identify mechanisms that are important in the etiology of autism in a subgroup of subjects. The biological impact of increased cytokines in autism children and their association with more impaired behaviors is intriguing and warrants further consideration. (Key Words: Immune Impairments)
Project 3 - Neurodevelopmental Toxicology
Description - The overall goal is to determine if exposure to environmental toxicants early in development contributes to the etiology of neurodevelopmental disorders. A related goal is to determine whether susceptibility to autoimmune disease increases the neurotoxicity of environmental contaminants and increases risk for developmental neurotoxicity.
Key Findings:
Our data indicate that developmental exposure to chronic low-levels of BDE-47 result in significant bioaccumulation in target organs, including brain, and is associated with early motor deficits in exposed neonates. (Key Words: Animal Models of Autism)
A study on the neurodevelopmental toxicity of autoantibodies and the effects on behavior in mice showed increased anxiety in mouse pups exposed during gestation to IgG-AU, consistent with our hypothesis that autoantibodies in the serum of mothers of autistic children may alter normal behavioral development. Our misoprostol study showed that there were no statistically significant effects between treatment groups in any of the parameters tested and does not support that early misoprostol exposure impairs normal brain development. (Key Words: Animal Models of Autism)
We identified mechanisms of developmental neurotoxicity of PCBs and PBDEs. (Key Words: Animal Models of Autism)
We showed how a non-coplanar PCB, PCB95, alters the structure and function of a major Ca2+ channel found in muscle and brain. PCB 95 has been found in significantly higher concentration is post-mortem brains from individuals with syndromic ASD than those obtained from unaffected controls. We found that developmental PCB exposure accelerated dendritic growth in untrained experimental animals when compared to untrained controls. The trained PCB-exposed animals, however, took longer to learn the water maze and showed reversal of dendritic growth in response to water-maze training, in contrast to controls that learning increases dendritic growth. The results are important because problems in dendritic growth and plasticity have previously been implicated in many neurodevelopmental disorders, including ASD. We also showed dysregulated expression of ryanodine receptors (RyR1 and RyR2) is a consequence of PCB exposure. We think PCBs are increasing the activity of these Ca2+ channels, which we know generate the signals needed for the extension and branching of dendrites. (Key Words: Animal Models of Autism)
