Objective:
The Center’s core purpose is to identify and address key emerging issues related to health impacts of early life environmental exposures on children. There is a growing body of research connecting exposures early in life to a number of prevalent and life threatening diseases for children. Moreover, it is important to understand these exposures as effects that begin to manifest in childhood may extend beyond, into adult life. Significant gaps remain in our understanding of the effects of early life exposures and of potential treatments or measures to reduce harmful exposures, especially in utero or in early childhood. Thus, the overarching goal of this Center is to conduct research that enables risk assessors, regulators, policy makers, caregivers, and other stakeholders to increase their capacity to implement immediate and long-term strategies to prevent environmentally induced diseases associated with environmental exposures, such as arsenic in children.
Progress Summary:
Major Activities
- Our synergistic, collaborative Center focused on the integration of all projects and cores. It remains productive and at the forefront of children’s environmental health research.
- Our established administrative structure supports our multidisciplinary research teams, and ensures clear, organized communications, budget management, fiscal planning, and regulatory compliance. The centralization of research activates is done through the Administrative, Research Integration and Career Development Core, and has led to enhanced productivity within the projects.
- The innovative new technologies and methodologies being used continue to enhance the promising research program of our Center. These include epigenetic analysis, precision gene expression arrays using NanoString, deep sequencing microbiome analysis, state-of-the-art immune profiling, and trace element measurement of biologic samples. These mechanisms have stimulated high impact biomedical research in children’s environmental health, both within our Center, and on a national and international scale.
- Our robust mentorship program, pairing early career investigators with recognized scientists, and leaders within our interdisciplinary team, remains a positive and effective element of our Center, and continues to further both the careers of our talented investigators and the Center’s research.
- The integrative Child Health team, led by prominent pediatric medical geneticist, Dr. John Moeschler, has continued to deepen connections among health care and public health providers, communities, parents, caregivers, and other key stakeholders to effectively reduce risk of environmental threats to children’s health.
- Our Community Outreach Translation Core (COTC) continues to partner with the Administrative Core, Child Health Specialist, New Hampshire Birth Cohort Study (NHBCS) participants, practice networks and public health partners to translate findings to stakeholders in clinical practices. The COTC continues to pilot a research study to examine the most effective ways to communicate research findings and risk, while observing behavior modification of key stakeholders—parents of children in New England.
- Our communication protocols include having our COTC Director, Dr. Carolyn Murray—a recognized and respected clinician—review and comment on all press releases and media communications before they are made public. This has greatly streamlined the Center’s messaging to maximize the effectiveness of our research translation.
Significant Results
Our well established and growing New Hampshire Birth Cohort Study (NHBCS) of maternal-infant dyads, continues to be of paramount importance to our research and Center. Building upon detailed exposure history, state-of-the-art biomarker measurements, and a rich collection of archived biospecimens obtained from this cohort, we are accelerating the pace of scientific advancement by leveraging additional Dartmouth resources, such as the Biorepository Core facility. Utilizing said facility as an archiving and retrieval mechanism for our precious biological samples, greatly increases the rate at which we can obtain data, and publish our findings. In this reporting period, our Center continued to refine database creation and management, providing researchers and trainees web access to data descriptions, which is vastly accelerating our scientific productivity, and will continue to be a focus in Year 4 of our Center grant. To advance the expansion of the Center’s research, we were awarded two successful Administrative Supplements, and continue to implement several pilot projects and sub-studies as well as major additions to the cohort study. Researchers within our cohesive Center continue to contribute important scientific findings via national and international conferences and publications, and communicate them to key stakeholders to reduce environmental threats to children and improve their lifelong health.
Key Outcomes/Achievements
Some of our Center’s key achievements over the past year are mentioned below, and highlighted within each core/project report.
- Project 1 published several papers on novel findings related to fetal and perinatal exposures and the stool microbiota of premature infants; the association of cesarean delivery and formula supplementation with the intestinal microbiome of 6-week old infants, and infant infection and respiratory outcomes in relation to in utero arsenic exposure. To advance the understanding of the role of diet and potential exposure to arsenic, Project 1 in collaboration with Project 2 has worked extensively in this reporting period to analyze dietary data and has several papers elucidating findings related to rice consumption and potential early life exposure to arsenic through rice-containing products.
- Dr. Margaret Karagas was an invited presenter at the National Academy of Sciences (NAS) Emerging Science for Environmental Health Decisions workshop “Environment and Health: What’s the Human Microbiome Have to Do with It?” in Washington, DC (January 2016).
- Dr. Wes Viles, a postdoctoral associate presented on “Entropy, statistical dependence, and network structure of the human infant gut microbiome” at the Pacific Symposium on Biocomputing in January 2016 on the Big Island of Hawaii, and again at the Microbiome Data Analysis in New York, New York in February 2016.
- Dr. Juliette Madan was an invited speaker at the "Altering the Microbiome: Can it Impact Health?" Biopharma Research Council Meeting co-sponsored by NIH/NCI on "Neonatal Microbiome: Exposures and Outcomes" (September 2015).
- Dr. Anne Hoen, a newly hired early career investigator within the Department of Epidemiology at the Geisel School of Medicine at Dartmouth, has research expertise in microbiome analysis and spatial modeling, and is working with Dr. Karagas on two recently awarded administrative supplements. These supplements are greatly enhancing our novel research on the microbiome and are propelling the project's findings, and the Center research as a whole.
- Dr. Anne Hoen and graduate student Sara Lundgren were invited to present at the Human Immunity and the Microbiome in Health and Disease Conference in Montreal, Canada (September 28-29, 2015).
- Dr. Jiang Gui presented a poster and abstract, in collaboration with Dr. Margaret Karagas, of the work, “A Two Step Variable Selection Procedure Using Prioritization of Interactions” followed by LASSO, for the NIEHS Statistical Approaches for Assessing Health Effects of Environmental Chemical Mixtures in Epidemiology Studies Workshop in Research Triangle Park, North Carolina, July 2015.
- Dr. Tracy Punshon, an early career investigator with Project 2 has been highly productive this reporting period investigating metal concentrations in the placenta in relation to other maternal and infant biomarkers. Dr. Punshon’s work in this area resulted in publications in Environmental Science & Technology and the Journal of Exposure Science and Environmental Epidemiology with several other papers published and underway.
- Project 2 postdoctoral associate Courtney C. Carignan developed expertise in arsenic exposure assessment and dietary epidemiology through her work with Drs. Cottingham and Gilbert-Diamond. Carignan led three manuscripts emerging from Aim 1 of this Project; the first appeared in Environmental Health Perspectivies in May 2015, the second in the Journal of Exposure Science and Environmental Epidemiology in November 2015, and a third was published this year in the Annals of Global Health.
- Dr. Susan Korrick of Project 2 is mentoring a post-doctoral fellow, Mahsa Yazdy, who is analyzing parallel data among another birth cohort (the New Bedford Cohort) with biomarkers of prenatal arsenic exposure. Collection of measures of cognitive and behavioral function through adolescence has been completed. Findings from analyses of this complete dataset will help inform analyses of the NHBCS neurobehavioral data.
- Faculty Development Investigator, and Project 2 co-leader, Dr. Diane Gilbert-Diamond is continuing to study how environmental factors influence early growth and obesity in children. She continues to co-mentor graduate student, Anala Gossai and has published several papers in this reporting period examining the relationship between in utero arsenic exposure and birth outcomes, gene-gene interactions in a statistical epistatis network of 12 candidate genes for obesity, gene-gene interactions and association with body mass index, and the effects of children’s food and beverage promotion on television to parents.
- Project 3 has continued to focus on the influence of environmental exposure on human epigenetic regulation, which has further fortified the strength between the NHBCS (PI: Karagas) and RICHS (PI: Marsit) birth cohorts.
- Dr. Benjamin Green, a second year postdoctoral fellow with Project 3 published a manuscript “The role of placental 11-beta hydroxysteroid dehydrogenase type 1 and type 2 methylation on gene expression and infant birth weight,“ in Biology of Reproduction and he was lead author on the arsenic EWAS study that was published in Environmental Health Perspectives on arsenic and placental DNA methylation in the NHBCS.
- Dr. Marsit, co-leader of Project 3, presented on “Readily Measurable Epigenetic Marks and Significance” at the U.S. EPA offices in Washington DC, for a workgroup considering epigenetic mechanisms in risk assessment. In addition, he presented seminars on Placental Epigenetics in Children’s Environmental Health to the Superfund Research Program at the University of Kentucky, at the Southern California Environmental Health Sciences Center at the University of Southern California, and at the Emory University Rollins School of Public Health.
- Dr. David Robbins of project 3 continues his work on an NIEHS Children’s Center administrative supplement to investigate sex differences in the Center’s data. He now is in the process of examining how arsenic speciation might contribute to some of the sexually dimorphic responses, and has published on a key development gene over the past year.
- Dr. Carolyn Murray, our COTC Director worked with the Dartmouth Neukon Center DALI (Digital Arts, Leadership and Innovation) lab to produce an educational tool for parents and the public on arsenic sources and consumption. This work was featured in the August 2015 NIEHS PEHP newsletter.
Future Activities:
During the next reporting period, we will continue to promote the discovery of new scientific findings through a multidisciplinary, integrated approach encompassing the spectrum from exposure, exposure biomarkers, response biomarkers, health outcomes and practice, and policy change of our interdisciplinary Center. Our focus continues to be identifying, and in turn reducing, adverse health consequences of environmental exposures in children. We will continue to strive to communicate new and emerging results to appropriate audiences in order to facilitate change in public health and clinical practice.
