Objective:
The main aim of this project is to determine the health effects of prenatal and postnatal exposures to individual pollutants, sources, and pollutant mixtures on somatic growth, cardiovascular risk (blood pressure, exercise tolerance) and cognition in children. The strength of chronic and acute effects of individual pollutants will vary by source and mixture, as well as the timing of prenatal and postnatal exposures. Increased vulnerability or susceptibility to pollution effects on these adverse health outcomes will also result from socioeconomic disparities, stress and violence, environmental tobacco smoke, and reduced maternal and child omega-3 fatty acid intake measured in the prenatal as well as postnatal periods. Our Advisory Committee recommended including asthma as an outcome, and evaluating genetic variants as sources for susceptibility.
Progress Summary:
During the past year we have continued to make excellent progress in each of the specific aims of this project. Our key accomplishments are summarized briefly below.
Focusing on early life outcomes and outcomes up through mid-childhood, we have analyzed associations of temporally-resolved and spatiotemporally resolved short-term and long term averages of individual pollutants and markers of traffic with a variety of outcomes of interest, including:
- Maternal and child metabolic and inflammatory outcomes
- Somatic growth
- Maternal and child blood pressure
- Cognition and behavior
- Respiratory illness in early life, lung function, wheeze and asthma
We began evaluation of longitudinal evaluation associations of fructose and pulmonary outcomes as first step in assessing this and other dietary factors as sources of vulnerability to pollution (collaboration with Michigan group). Genome-wide measurement of DNA methylation at birth and in early life will offer many opportunities to explore pathways through which pollution influences outcomes.
We are assessing the relation of measures of socioeconomic status to green space, distance to roadway, sources of air pollution, and levels of air pollution. We apply this work to consideration of individual, family, and neighborhood markers for socioeconomic status and urbanicity as potential confounders or effect modifiers of associations of pollution with Project Viva outcomes. In collaboration with investigators from all projects we have collaborated on definition of pollutant, pollutant component, and weather clusters. We have begun to look at effects of pollution sources and mixtures on Project Viva outcomes.
We have estimated spatio-temporally resolved PM2.5 with a 1 km spatial resolution and spatio-temporally resolved temperature with an address-level resolution, to assess their relationships with our childhood outcomes.
Future Activities:
Our plans for the next year in Project Viva are to complete and submit additional papers on prenatal individual pollutant exposures and early life outcomes, and evaluate the effects of pollution sources and mixtures. We will complete the longitudinal exposure and health outcome data to evaluate individual pollutant associations with repeated measures outcomes up through age 7. With investigators from all projects we have collaborated on definition of pollutant, pollutant component, and weather clusters. We will expand investigation of effects of pollution sources and mixtures on Project Viva outcomes. We will continue to assess sources of vulnerability and susceptibility to individual pollutants, pollution sources and mixtures. We will also continue to evaluate effect modification by family and neighborhood socioeconomic status, nutrition, and genes. Our pathway analyses evaluating the role of epigenetic and biomarker outcomes will continue, and we will complete our GXE work in the CAMP study.
