Objective:

We aim to improve toxicity testing for novel proteins in animals by studying the role in the gut of a protein biomarker, thymic stromal lymphopoietin (TSLP), with a central role in allergic inflammation.

Progress Summary:

In this project period, we have undertaken studies to determine the role of TSLP in allergy and tolerance in the gastrointestinal tract. We have determined that TSLP is necessary for a model of allergic inflammation localized to the gastrointestinal tract, but is not required for primary allergic sensitization or immune tolerance generated via the intestinal mucosa. Our mechanistic studies show that TSLP is required for the induction of pathogenic Th2-producing T cells, and that the target of action of TSLP is on the memory T cell. Therefore, we conclude that TSLP plays an important role in the amplification of allergic sensitization into allergic disease in the gastrointestinal tract.  

Future Activities:

  1. Assessment of the regulation of TSLP expression by human intestinal epithelial cells.
  2. Assessment of the role of TSLP as an initiator of allergic sensitization to food proteins.
