Objective:

1) Determine the NOAEL and the LOAEL of food proteins with high vs. low/no allergenic potential in a mouse model.

2) Determine the immune, clinical and physiological responses to isolated pure food proteins with high vs. low/no allergenic potential in this model.

Progress Summary:

(1) Determination of allergenicity and oral elicitation threshold doses (LOAEL and NOAEL) for hazelnut protein prepared by our laboratory;
 
(2) Evaluation of the allergenicity of sesame seed protein from two sources (our lab prepared sesame seed protein and obtained it from the Greer Lab) in the TS/OE model;
 
(3) Determination of oral elicitation threshold doses (LOAEL and NOAEL) for these two sources of sesame seed protein in TS/OE model; and
 
(4) Estimation of human equivalent threshold doses for sesame proteins from both sources.
 

Future Activities:

We plan to evaluate the allergenicity of purified protein ovalbumin, establish oral-dose response curves and percent responder curves, and estimate oral elicitation threshold doses (LOAEL and NOAEL) in TS/OE mouse model. Based on the mouse model data and using published methods, we will estimate the human equivalent threshold doses for ovalbumin. Subject to availability of adequate funds, we plan to evaluate allergenicity of another purified protein, rubisco, in this mouse model. We plan to submit a manuscript for publication and present an abstract for presentation during the American Association of Immunologists Annual Meeting to be held in May 2014.
