This study evaluated the percutaneous absorption of dimethylcyclosiloxane fluid. Four tissue samples from each of 3 human cadavers was used for these in vitro studies. Five ul doses containing 1.5 uCi radiolabelled dimethylcyclosiloxane fluid were applied to each 1 square centimeter tissue sample. Skin integrity was also evaluated by histopathological examination. Histopathological examination indicated no lesions on the tissue sections. After 24 hours, the total percutaneous absorption (skin compartment and amount absorbed through the skin) was 0.29 percent, 0.47 percent, and 4.4 percent of the applied amount. Greater than 80 percent of the total compound recovered was found in the air traps confirming the relative volatility of this compound. Under conditions of this experiment, the compound was found to volatilize prior to significant absorption. |
