Description:

The purpose of this grant is to establish in an unambiguous way, the catalytic properties of the cytochrome P4501A protein in fish. This protein is induced (i.e., increased in amount) in many organs of fish exposed to polynuclear aromatic hydrocarbons and planar halogenated aromatic hydrocarbons in the environment. This induction is the most robust indicator (biomarker) of exposure to such compounds. Knowing the exogenous and endogenous substrates metabolized by the CYP1A1 will greatly improve the interpretation of the biological significance of that induction. To obtain this information, the gene for CYP1A1 was cloned from the marine fish scup, and the gene was then incorporated into a cell line that does not express other P450s. The ability of the scup CYP1A1 made in these cells to metabolize a variety of candidate and diagnostic substrates is being examined, without the interference by other P450s. This is difficult, if not impossible to do, with preparations of animal tissues, which express other P450s in addition to CYP1A1.
