Objective:
This is a proposal to establish a Children's Environmental Health and Disease Prevention Research Center (P01) at the University of Illinois. The Center would expand and extend research initiated under our currently funded Formative Children's Center (P20). The primary goal is to use a multidisciplinary approach to address critical gaps in our knowledge about the impact of exposure to endocrine disrupting chemicals on child development. We will investigate how endocrine disrupters (phthalates; BPA) interact with diets high in saturated fat to impact neurological and reproductive function. The impact of exposures during two critical developmental periods - prenatal and adolescent - will be assessed. At the heart of the research effort will be two human cohorts that together make up Project 1- a prenatal exposure cohort which involves expansion of a prospective pre-birth cohort that is currently underway in Urbana, Illinois and an adolescent exposure cohort to be studied in the context of a long-standing prospective birth cohort study currently underway in New Bedford, Massachusetts. The Center will also include two laboratory rodent studies (Projects 2 and Project 3) that will model the timing of exposures assessed in the two human cohorts. The laboratory studies will explore mechanisms of action and will also help to identify neurological and reproductive outcomes that will be important to assess in longitudinal follow-ups of infants and children in the human cohorts. Central to all three research projects are three key aims: (1) assessment of exposure during two critical periods (prenatal and adolescent), (2) investigation of joint effects of exposure and diet, and (3) investigation of the role oxidative stress and inflammation in mediating outcomes. Projects 2 and 3 will use a phthalate mixture that is based on the pattern of phthalate metabolites found in human urine from Project 1. Additionally, we will actively involve and mentor junior investigators new to children's environmental health research, and we will develop and evaluate innovative strategies to communicate our research findings to the parents, childcare providers and healthcare providers through our Community Outreach and Translation Core (COTC). Center activities will be coordinated through an Administrative Core.
