Description:

The Cytochrome P450 array detoxifies many chemicals by catalyzing the conversion of mostly hydrophobic chemicals into more hydrophilic forms that can subsequently be excreted by the body. Human genetic variation in the genes for these enzymes produces wide variations in the ability of individuals to detoxify chemicals of environmental and occupational concern. We have previously shown that differential expression of CYPIAI gene predisposed individuals in some families to lung cancer caused by smoking. Here we review the current state of the art regarding differential P450 expression and predisposition to cancers caused by several environmental toxins as well as toxicants in the workplace. Additionally, implications of P450 gene diversity for drug design and emergency room treatment of overdoses is considered.
