Typically in vitro hazard assessments for the identification of endocrine disrupting compounds (EDCs), including those outlined in the EDSTAC Tier 1 Screening (T1S) protocols, utilize mammalian receptors. However, there is uncertainty concerning differences that may exist in the binding affinities of toxicants for steroid receptors from non-mammalian species. To test the hypothesis that EDCs bind steroid receptors from mammalian and non-mammalian vertebrates and invertebrates with similar affinity, tissues were collected from five different species for cDNA library construction and isolation of steroid receptor sequences. The current work reports the isolation and sequencing of both an estrogen receptor and an androgen receptor from the fathead minnow (Pimephales promelas). The fathead minnow steroid hormone receptor sequences, and eventually other isolated receptors, will be used to generate recombinant receptor proteins for binding and in vitro assays for cross species extrapolation studies. These studies will facilitate the identification of EDCs that affect many species and ultimately impact future risk assessment protocols.
