XENOBIOTIC METHODS DEVELOPMENT FOR HUMAN EXPOSURE ASSESSMENT RESEARCH

Impact/Purpose:
The objective of this task is to develop state-of-the-art methods for measuring xenobiotic compounds, to include the isolation of the analyte from the appropriate matrix (extraction), preconcentration (typically sorbent-based), and analysis via GC/MS and/or LC/MS. Once established, these methods will be applied in small scale pilot studies or demonstration projects. Particular emphasis will be placed on methods which are readily transferable to other laboratories, including those within the Human Exposure and Atmospheric Sciences Division (HEASD), the National Exposure Research Laboratory (NERL), other EPA Laboratories, Program Offices, Regions, and academic institutions.
Specific objectives of this task include the following:
1) Development of GC/MS and LC/MS methods for the measurement of key xenobiotic compounds and their metabolites (to include the pyrethroid pesticides, perfluorinated organic compounds, and the BFRs) in relevant environmental and biological matrices.
2) Development of efficient low cost methods for the extraction and clean up of these compounds collected from relevant matrices.
3) Determination of xenobiotic compound and metabolite concentrations in samples derived from laboratory and field monitoring studies to help assess exposures and evaluate associated risks.
Description:
Biomarkers from blood, breath, urine, and other physiological matrices can provide useful information regarding exposures to environmental pollutants. Once developed and applied appropriately, specific and sensitive methods can often provide definitive data identifying the various sources of exposure; the magnitude of exposure; and the uptake, elimination, and body burden of specific compounds. These data can be essential for the appropriate evaluation of the risks incurred with particular exposures.
This task focuses on the development and application of gas chromatography/mass spectrometry (GC/MS) and liquid chromatography/mass spectrometry (LC/MS) based biomarker methods for use in exposure assessment research. Emphasis will be placed on the development of methods to investigate exposures to current- and future-use pesticides, key perfluorinated organic compounds (e.g., perfluorooctane sulfonate [PFOS] and perfluorooctanoic acid [PFOA]), brominated fire retardants (BFR) and other high priority compounds identified by State, Regional, and Program Offices.

Project Information:
Progress :A Quality Assurance Project Plan (QAPP) covering all of the work under this task has been completed.A new GC/MS system, to accelerate the pace of this research, has been brought in house to replace an older instrument. A variety of derivatization techniques have been explored for the GC/MS analysis of the key pyrethroid metabolite 3-phenoxybenzoic acid. These methods are undergoing characterization with regard to their efficiency, linearity, and sensitivity.
House dust samples (N=112) from the Children's Total Exposure to Persistent Pesticides and Other Persistent Organic Pollutants (CTEPP) program have been sieved to < 150 um in preparation for research designed to characterize xenobiotic materials in this matrix.
GC/MS methods for the most common perfluorinated telomer alcohols have been developed along with a sorbent-based solid phase extraction method for the clean up of environmental extracts. A manuscript covering the distribution of the various telomer alcohols in house dust is in preparation.
Preliminary experiments with representative brominated fire retardants indicate that accelerated solvent extraction may be an appropriate technique for the isolation of these materials from house dust.
The new LC/MS system (an Applied Biosystems API 3000) has been installed and is now fully operational. A method for the measurement of PFOS in blood serum has been adapted from various industry and literature sources and further refinements are now underway to obtain a lower limit of detection and to extend this technique to environmental matrices.
GC/MS method for perfluoro-telomer alcohols delivered to Eric Weber (NERL-Athens) and Matthew Henderson (University of Georgia). Client feedback is that the method is currently used and was very helpful.

Relevance :This research provides methods and data which will enable the National Exposure Research Laboratory (NERL) and the Office of Prevention, Pesticides & Toxic Substances (OPPTS) to develop a more complete understanding of residential exposures to current use pesticides and other key xenobiotic compounds, thereby reducing uncertainties associated with the reliance on default modeling assumptions. Specifically, this effort provides consistent and scientifically sound methods and approaches for measuring exposure; helps determine the major factors that contribute to variability in exposure; and, supports current and prospective measurement studies designed to determine aggregate and cumulative exposures for vulnerable subpopulations.
