Description:

Glucocorticoids are important regulators of embryonic growth and development. hese effects are mediated through glucocorticoid receptors (GR) which bind to glucocorticoid response elements upstream of regulated genes. his study examines the expression of GR and GR mRNA in embryonic secondary palate from visceral arch stage through fusion. mbryonic rat (gestation day (GD) 12) and mouse (GD 10, 12, 13, 14, and 15) were examined by in situ hybridization with 35S-riboprobe prepared from rat GR CDNA, and GR was localized immunohistochemically with the monoclonal antibody BUGR-2. isceral arches express GR MRNA uniformly across first and second arch mesenchyme, but no GR was detected immunohistochemically. n GD 12, MRNA localized predominantly to the region of palatal shelf outgrowth, but GR was not detected in palates or adjacent tissues. n GD 13 MRNA appeared in oral epithelia and adjacent mesenchyme and GR was detected at low levels. y GD 14 GR was strongly expressed in central mesenchyme and adjacent to oral epithelium and the MRNA was abundant in oral and medial mesenchyme, particularly in regions of bone formation.
