Impact/Purpose:

The purpose of these slides is to provide an overview and training of how to use the CompTox Chemicals Dashboard to examine and use ToxCast bioactivity data.

Description:

New approach methodologies (NAMs) for hazard can be used to inform biological, weight-of-evidence questions as well as risk-related questions for a given substance or set of substances. In this training, a generic workflow using the CompTox Chemicals Dashboard as a data viewer and download utility is presented. This workflow includes consideration of the domain of screening applicability based on physicochemical properties, consideration of bioactivity and cytotoxicity information, followed by integration with a high-throughput toxicokinetic approach to in vitro to in vivo extrapolation so that a bioactivity:exposure ratio (BER) can be estimated to indicate relative priority. In a first vignette, evidence to support relevant adverse outcome pathways for the discontinued drug, troglitazone, is examined. In a second vignette, consideration of endocrine bioactivity and relative priority via a BER is demonstrated. Trainees should gain a practical sense of how to access the necessary information to utilize NAMs to inform hazard prediction for weight-of-evidence evaluations, screening and prioritization, and point-of-departure prediction for a NAM-based safety assessment.
