Wambaugh, J. 20181030 - What Doesn’t for Extrapolation to In Vivo Exposures? Presented at RTP Drug Metabolism Discussion Group, Research Triangle Park, NC, October 30, 2018. https://doi.org/10.23645/epacomptox.7347389

Impact/Purpose:

This is a presentation on in vitro-in vivo extrapolation to the RTP Drug Metabolism Discussion group.

Description:

We would like to know more about the risk posed by thousands of chemicals in the environment – which ones should we start with? High throughput toxicokinetics (HTTK) is an example of new approach methodologies (NAMs) that are being evaluated through comparison between in vitro predictions and in vivo measurements of both plasma concentrations and doses associated with the onset of effects. Comparison between HTTK predicted time course concentrations in plasma and in vivo data indicate that some properties (e.g. average and maximum concentration) can be predicted with confidence. NAMs for TK allow risk-based prioritization of large numbers of chemicals.
