Impact/Purpose:

Thyroid hormones (THs) are essential for normal vertebrate development and the model organisms used in assessing the TH disrupting potential of chemicals are often the early development stages which have limited available tissue mass or plasma volume. This work describes sensitive analytical methods that can quantify TH levels in small or limited tissue samples. TH concentrations in plasma and/or tissue are critical measures in describing Adverse Outcome Pathways (AOPs) of the thyroid-axis. The information gained from this methodology can provide a sound scientific basis for supporting decision processes within EPA, for example OSCP’s EDSP, for evaluating chemicals for thyroid hormone disrupting potential.

Description:

This poster presentation will describe analytical chemistry methods for measuring thyroid hormones and related precursors and metabolites in very small tissue or plasma samples. These methods are amenable to measure thyroid hormones in amphibian tadpoles or small mammals used as model organisms for investigating the effects of chemicals on thyroid hormone-dependent developmental processes, or in any instance where tissue or blood samples are limited. The determination of thyroid hormones in these matrices provides a greater understanding of how exposure to chemicals disrupt normal thyroid hormone levels and can lead to adverse effects.
