Description:

We have measured the levels of amplification of oncogenes and tumor marker genes or other genes of interest in nine human lung tumor cell lines in comparison to normal human bronchial epithelial cells or normal blood lymphocytes to test the hypothesis that aberrant amplification of marker genes may be related to their proximity on the human genome to oncogenes similarly amplified during the development of malignancy. ix pairs of comapping genes and an additional unpairad oncogene were used for analysis: the oncogenes MYGL and BLYM, KRASIP and prolactin; KRASlP and chronic gonadotropin, alpha chain; MYB and chorionic gonadotropin, alpha chain; MYC and thyroglobulin; F0S and creatine kinase, brain form, and MUGN. f 31 gene pairs tested, amplification was observed in three cases. n two out of those three, co-amplfication was observed in the MYG gene and the closely mapping thyroglobul in gene. hese observations provide evidence that genetic alterations in tumors may involve more than one closely mapping gene and may provide a basis for coordinate deregulation of these genes. Preliminary field inversion gel electrophoretic analyses indicate that MYC and thyroglobulin may lie within 1,000 kilobase pairs of each other on the genome, providing an estimate of the size of amplification events occurring in tumor cell lines. In addition we have characterized a number of new tumor cell lines cytogenetically and have adapted semi-automated techniques for banding of mitotic chromosomes and for the detection of tumor marker and oncogene expression in tissue samples. hese techniques will be useful in future analyses of coordinated deregulation of genes in actual tumors rather than in tumor cell culture.
