Impact/Purpose:

This presentation will illustrate how toxicogenomics in combination with traditional toxicology approaches has been used to identify key events in a mode of action and how these data could be used to inform a cumulative risk assessment.

Description:

Toxicogenomic data, in this caser transcriptional profiling, has been valuable in providing a deeper understanding of the cellular and molecular pathways that are altered in response to environmental stressors including chemicals. This knowledge can be used effectively in the development of a risk assessment. For example, an important component of the risk characterization process is identifying and characterizing, in an organized manner, the key biological changes that describe a biologically plausible mode of action for the development of an adverse health effect. This is a critical feature of cancer risk assessment as described in the US EPA’s Guidelines for Carcinogen Risk (2005). Toxicogenomics is also a valuable approach for the identification of key events and for describing their molecular characteristics. Once a mode of toxic or carcinogenic action is described, toxicogenomics can be used to identify common altered pathways among a group of chemicals. Describing a common mode of action for a series of chemicals is critical for developing a cumulative risk assessment as described in the Food Quality and Protection Act (1996). This presentation will illustrate how toxicogenomics in combination with traditional toxicology approaches has been used to identify key events in a mode of action and how these data could be used to inform a cumulative risk assessment.
