Description:

This manuscript provides an overview of the use of data from toxicology and epidemiology studies for neurotoxicity risk assessment. Parameters such as the use of subjects, study designs, exposures, and measured outcomes are compared and contrasted. The main concern for use of data from toxicology studies is extrapolation, and that for use of epidemiology data is attribution. The most compelling evidence for establishing causation between exposure and outcome is consistency of reports across a number of studies, a principle at odds with the selection of a single critical study to provide a point of departure for risk assessment. The manuscript presents approaches to evaluate and improve concordance across species and consistency across studies. Arguments are illustrated with examples of recently published work. The impact of this manuscript could be to serve as a guide to evaluating toxicology and epidemiology data for risk assessment, and to facilitate the use of the entire body of available evidence, rather than a single critical study, to establish a point of departure for neurotoxicity risk assessment.
