Description:
The National Toxicology Program Center for the Evaluation of Risks to Human Reproduction (NTP-CERHR) convened an expert panel in May 2004 to evaluate acrylamide. The report of the expert panel, prepared in accordance with CERHR Guidelines, provides a detailed summary of all published information about the chemistry of acrylamide, its use and human exposure (general and occupational), general toxicology and biological effects including toxicokinetics and metabolism, genetic toxicity (in somatic and germ cells), carcinogenicity, developmental toxicity, and reproductive toxicity (male and female). Each section includes strengths/weaknesses of available studies and their utility for the CERHR evaluation process. The report concludes that acrylamide is of negligible concern in humans for adverse reproductive and developmental effects and of minimal concern for heritable effects for exposures in the general population, whereas it is of some concern for these outcomes for exposures in occupational settings, with consideration given to potential for neurotoxicity at similar exposure levels. Critical data needs include better definition of occupational exposure, better understanding of potentially susceptible subpopulations based on metabolic genotypes including CYP2E1 polymorphisms, a better definition of dose-response for heritable alterations (preferably using direct assays of germ cells), and a more definitive epidemiology study of occupationally exposed men.
