Cell culture models are often used in mechanistic studies of toxicant action. However, one area of uncertainty is the extrapolation of dose from the in vitro model to the in vivo tissue. A common assumption of in vitro studies is that media concentration is a predictive marker of tissue concentration. This assumption may not hold true for lipophilic chemicals. This manuscript characterized the relationship between chemical concentration in the media and final tissue dose of the toxicant PBDE-47 in a neuronal cell culture. For this lipophilic compound, there was a large magnification (100-fold) of the dose accumulated in the cells compared with the dose applied in the media. In addition, accumulation was affected by the composition of the media. Thus, the concentration of a compound in the exposure solution may not be the appropriate dose metric for in vitro studies, and accurate information on the concentration of a compound in cells should be determined experimentally.
