Impact/Purpose:

Chemical safety decisions can be hindered by the lack of ready-access to data, tools, and models. Decision support tools are needed to support chemical safety decisions, where case study examples can build the confidence needed for regulatory acceptance. The Adverse Outcome Pathway (AOP) framework helps to organize existing mechanistic information and contributes to New Approach Methodologies (NAMs). The EPA AOP Database (AOP-DB) was created because automatic and systematic parsing of AOP-Wiki data is challenging especially for non-technical users. The AOP-DB was developed to assist in the mechanistic characterization of AOP data for both hypothesis generation and case study development, and to provide the user the ability to take a “deep-dive” into AOP metadata, providing a systems-level overview of the biological context of AOPs, identifying associations with chemical, assay and disease endpoints to expedite predictive toxicology efforts. The EPA AOP-DB is a data query and retrieval tool, the first tool of its kind to make AOP and related meta-data easily accessible and machine-actionable. The 2024 iteration of the AOP-DB, version 3, will include the planned interoperability with a variety of EPA and EPA-partner data, models, and tools. Three critical areas of data, tool, and model integration have been identified through user, partner and stakeholder group discussions: 1. Prototype Chemical Identification and Ranking; 2. EcoToxicology Model Population Predictors; and 3. Human Population-level Susceptibility.
