Abstract |
The effects of certain reputedly non-genotoxic agents on cholesterol and DNA synthesis were investigated in cultured primary rat hepatocytes and in liver slices. Hepatocytes in culture were incubated for 48, 60, and 72 hours with one of the following chemicals: chloroform (CHCl3); 1,1,2-trichloroethane (TCE); trichloroacetic acid (TCA); dichloroacetic acid (DCA); monochloroacetic acid (MCA); di(2-ethylhexyl)phthalate (DEHP); mono(ethylhexyl)phthalate (MEHP); phenobarbital (PB); epidermal growth factor (EGF); dimethylsulfoxide (DMSO). All test chemicals stimulated cholesterol and DNA synthesis in the cultures. However, the induction of DNA synthesis in hepatocytes treated with DCA became apparent by sixty hours. |
