Abstract The purpose of this study was to determine the potential irritant effects of PyAOP on the eyes of rabbits. The test article was instilled once into the conjunctiva sac of the right eye of each New Zealand white rabbit as a packed volume of 0.1 ml/eye. Upon instillation, each eye was held closed for approximately 1 second to prevent loss of material. The left eye remained untreated and served as the control. Both eyes of all animals were examined and scored for ocular irritation prior to dosing and 1 hour after dosing. Ocular irritation scores at 1 hour post-dose included sluggish reaction of iris to light, beefy red conjunctivae, swelling of eye lids and discharge. Based on mortality observed for five animals, positive ocular scores, and as suggested by Calvert's Department of laboratory and Animal Medicine, the study Director requested the remaining animal be euthanized on Day 2. Based on the observations made in this study, PyAOP was determined to be a very severe irritant (Class 7) according to the Modified Kay and Calandra Interpretation of Eye Irritation Test (Table 3), as well as ocularly toxic when dosed as received.
