Abstract | Bis(2-ethylhexyl) phthalate (CAS No. 117-81-7) was evaluated for subchronic toxicity. The test substance was administered in the diet ad libitum to rats (10/sex/group) at doses of 0, 1,000, 4,000, 12,500, and 25,000 ppm for 13 weeks. There were no deaths. Relative to controls, males in the 1,000 ppm group had significantly increased liver weight and females had significantly increased kidney weights. At 4,000 ppm, there were low red cell counts (males), slight hepatocellular enlargement (males), increased liver weights (males and females), increased blood urea nitrogen levels (males), and increased kidney weights (males and females). Rats treated with 12,500 ppm had urine stains, mild anemia, kidney and liver changes (increase in weight and hepatocellular enlargement as compared to controls), and low body weight (females only). Low body weight gain, urine stains, mild anemia and microscopic changes of the pituitary gland, adrenal cortex, kidney and liver (increase in weight and hepatocellular enlargement as compared to controls), and testes/epididymides (decrease in weight and increase in atrophy and aspermia as compared to controls) were noted in the 25,000 ppm group. | |||||||||||
