Abstract |
Tetraethylenepentamine (TEPA) was evaluated for potential mutagenic activity with a battery of three in vitro tests, which were: the Chinese Hamster Ovary (CHO) Uutation test, the Sister Chromatid Exchange (SCE) test and an assay for induction of Unscheduled DNA Synthesis (UDS) in rat liver cells. The results indicated that the sample of TEPA produced a significant effect on primary DNA damagge assessed in the ECE and UDS tests but TEPA did not increase the frequency of gene mutations in the CHO Mutation test. The observation of a positive response in two of the three tests suggested that the complex mixture containing TEPA should be considered a weakly active mutagenic agent for mammalian cells in vitro. Because this sanple was a mixture which contained numerous other components, the present tests cannot discriminate whether TEPA itself or one or more components produced the positive responses. These results should be considered an indication of the potential activity of this material and they suggest the need for other tests to investigate the biological significance of the observations of this study. |
