Abstract |
Interactions of the metals cadmium and selenium with various biologically important substrates were studied by nuclear magnetic resonance (NMR) spectroscopy. Cadmium-113 NMR was used for a critical examination of three metalloproteins: concanavalin A, bovine superoxide dismutase and carboxypeptidase A. The NMR parameters of selenium-77 were investigated, with a view to using this nucleus as a probe of active site sulfydryl groups in proteins. Several advances in NMR instrumentation were developed to further the aims of this project. One is a unique NMR probe, capable of spinning large (18 mm) NMR tubes, decoupling at any frequency, and observing any NMR-active nuclei. A decoupler modification, 'Chirp' decoupling, was developed. This modification allows good experimental results with approximately 1/10 the power required without modification. |
