The ability of oleylamine to induce specific locus mutations at the TK locus in cultured L5178Y mouse lymphoma cells (Mouse Lymphoma Mutagenesis Assay) was evaluated both in the presence and absence of metabolic activation from Aroclor-induced rat liver S9 fraction. Based on preliminary toxicity tests, 10 non-activated cultures treated with 0.13, 0.18, 0.24, 0.32, 0.42, 0.56, 0.75, 1.0, 1.3, or 1.8 nl/ml produced a range of 3-110% total growth. Eight activated cultures treated with 1.3, 1.8, 2.4, 3.2, 4.2, 5.6, 7.5, or 10 nl/ml produced a range of 6-106% total growth. Mutant frequencies in all treated cultures were less than 2 times that of the solvent (DMSO, acetone) controls, either with or without metabolic activation. |
