Abstract
The report pertains to 4-methoxy-1-naphthalenol (synonym: 4-methoxy-1-naphthol) (CAS No. 84-85-5) and is being submitted because of effects observed in a study conducted by multiple exposure routes. The title of the report being submitted is: 'Basic Toxicity of 4-Methoxy- 1-naphthol'. The latter report is being identified as a study involving other than human effects (Unit II.B.2.b of CAP Agreement). Groups of five rats were fed 1.0 % (925 mg/kg/day) or 0.1 % (100 mg/kg/day) of the test compound in the diet for twelve days. Exposure at the high-dose level decreased feed intake and reduced weight gain. Abnormalities in the high-dose group included: a great increase in absolute and relative spleen weights, enlarged spleens, and splenic congestion not associated with any lymphoid or reticuloeodothelial effect- slight decrease in hemoglobin and hematocrit, and moderate anisocytosis, poikilocytosis and polychromasia- and moderate increase in absolute and relative polymorphonuclear leukocytes. A slight increase in polymorphonuclear leukocytes and enlarged spleens with splenic congestion were also seen in the low-dose group. This chemical was also a strong skin and eye irritant and a weak to potent skin sensitizer in 9 of 10 animals tested.
