The test material was evaluated for genetic activity in a microbial assay (Ames, et al., MUTATION RESEARCH 31:347, 1975) with and without the addition of mammalian metabolic activation. Salmonella typhimurium Str. TA-1535, TA-1537, TA-1538, TA-98, and TA-100 were used as indicator organisms. Dimethy1sulfoxide, absolute ethanol, and sterile physiological saline extracts of the test material prepared by continuous rotary shaking (150 rpm/37C/24 hours) in accordance with the U.S.P. Class VI Plastics Method for biological testing of solid materials, were each introduced into the test system at concentrations of extract ranging from 50 to 3.12pl. A Spot Plate Test against each of the indicator organisms was conducted using the extracts directly on the surface of the agar test plates. No evidence of genetic activity was observed. |
