Abstract |
The document provides a series of protocols by which a trained technician can conduct studies on the transfer of genetic information by conjugation or transduction in soil, with emphasis on bacteria containing recombinant DNA. The level of the document is geared to technicians with some background and experience in standard laboratory methods used in microbiology but who have limited knowledge of, and experience in, bacterial genetics, molecular biology, and microbial ecology, especially in soil. Many of the specific techniques described in the document for studying gene transfer in soil were developed by the authors during the past few years. The primary motivation for the studies was the need for information on the survival of, and gene transfer by, genetically engineered bacteria that could be used in risk assessment of the release of genetically engineered microorganisms to the environment. |
