Abstract
The number of new and old chemical compounds to which the human race is exposed has quickly become astronomical. Methods of risk assessment have been slow to develop. Industry, government and academia have had to scrupulously examine the benefits-to-cost ratio of these studies. The limiting factors appear to be the questions of extrapolation and the compatibility of different metabolic processes in different species. The chapter discusses (1) pharmacokinetics, (2) model development, (3) flow-limited assumptions, (4) membrane-limited assumption, (5) micropharmacokinetic physiologic models and their implementation.
