Abstract
SAP 011 (containing calcium naphthenate, purity not reported) was evaluated for reproductive toxicity in 12 male rabbits receiving once daily occluded applications of 2 ml undiluted SAP oil, 6 hours/day, 5 days/week for 10 weeks followed by mating. Control animals received identical applications of the 'carrier oil' component present in SAP 011. No systemic toxicity, application site toxicity, or statistically significant changes in body weights were observed in male animals during the 10 week exposure or 12 week post mating periods. Additionally, no significant differences in testes weights were observed in male animals sacrificed after either period. The treatment did not affect the number of males achieving one viable litter, two viable litters, or two pregnancies. The number of implantations, pre- and post- implantation losses, and viable fetuses were not significantly different in females mated with exposed males, compared to those mated with unexposed males. The investigators reported that there were no macroscopic or microscopic pathological findings in the male reproductive tract that could be related to SAP 011 exposure.
