Absorption and metabolism of 2-chlorotoluene was examined in four female and four male Sprague-Dawley albino rats administered a single, oral 1 mg/kg bw dose of (U-ring-14C)2-chlorotoluene by gavage. Following administration, 2-chlorotoluene was rapidly absorbed; near complete elimination of the dose occurred within four days. No significant sex-related differences in metabolism of the test compound were found between males and females. A total of 85-92%, 5-8%, and 1-4% of the applied 14C were excreted in the urine, feces, and expired air, respectively. The primary urinary metabolites were 2-chlorobenzoic acid, a glucuronide of 2- chlorobenzyl alcohol, and a mercapturic acid, representing 20-23, 35-42, and 21-28% of the urinary 14C, respectively. |
