Abstract
Isopropanol (CAS No. 67-63-0) was tested for reproductive toxicity. The test substance was administered orally in the drinking water at 0, 0.5, 1.0, and 2.0% to male (10/group) and female (30/group) rats. The females were treated for 21 days before mating and continued receiving treatment throughout the study. After a 70-day treatment period, each male was paired with three females at the same treatment level; of the three females, one was sacrificed on day 19 of gestation and the two were allowed to litter. As compared to controls, at 2.0%, decreases in food and water intake and body weight, and increases in kidney and liver weights were noted in both sexes, and there were decreases in pup weight gain and survival. The fetuses of the sacrificed females in the highest group had whole body edema and slightly reduced body weight as compared to controls. The surviving pups had dose-related increases in relative liver weights. The NOEL for reproductive effects was 1.0%.
