Abstract
A study evaluated the potential toxicity of dimethylcyclosiloxane fluid following oral administration. Five groups of 8 male and 8 female Sprague Dawley rats were administered 0, 25, 100, 400, or 1600 mg dimethylcyclosiloxane fluid/kg/day by gavage for 14 days. All animals were observed daily for general appearance, behavioral abnormalities, signs of local or systemic toxicity, and mortality and weighed prior to study initiation, on Day 7, and prior to terminal sacrifice. All animals surviving to Day 14 were sacrificed, necropsied, and their livers removed and weighed. No treatment related changes in mortality, general appearance, behavior, or body weight and no overt signs of toxicity were reported. Statistically significant increases in absolute and relative liver weight were observed in the 1600 mg/kg females and in the 400 and 1600 mg/kg males.
