Abstract
This study evaluated the potential adverse effects of 100 and 350 cs polydimethylsiloxane (PDMS) fluid on the gonads and accessory sex organs of rabbits. Undiluted test substance was subcutaneously injected at a dose of 2.0 g/kg once daily in New Zealand rabbits (10 males and 10 females per dose group) for 20 days. An untreated control group and a positive control group in which DC555 fluid was injected at a dose of 50 mg/kg (5 males and 5 females each group) were also employed. At the end of the study, gross necropsies and microscopic examinations were performed. No deaths, untoward effects or gross pathologic alterations were reported. Thyroid gland weights and thyroid gland to body weight ratios were significantly increased in animals treated with 100 and 350 cs PDMS. Spermatogenic depression was reported in 3 positive controls and 2 animal for each cs PDMS fluid.
