Description:
This highly integrated study will test the hypothesis that the accumulation of the major heat inducible stress proteins, stress-70 and chaperonin (cpn60), reflect contaminant-induced damage to tissues and subcellular compartments. Two polyaromatic hydrocarbons, benzo(a)pyrene and beta-naphthoflavone, and copper will be investigated for their effect on individuals, cellular and subcellular levels of biological organization using molecular biological, physiological and comparative pathobiological approaches. These hallmark features of response will be linked to induction and accumulation of stress proteins in rainbow trout (Oncorhynchus mykiss) and medaka (Oryzias latipes). After testing the relationship in laboratory studies under conditions of acute, acute with recovery, and chronic exposures to various life stages, we shall apply the most relevant methods to evaluate a copper contaminated field site and an upstream reference site inhabited by rainbow trout. The latter will allow us to account for potential confounding results in field versus laboratory situations. Results will identify target tissues of toxicity and will be used to better interpret meaning of stress protein responses.Publications and Presentations:
