Objective:

The effects on children’s exposures of recent changes in pesticide uses, such as those resulting from the federally mandated phase-outs of residential use of chlorpyrifos and diazinon and the increased use of alternative pesticides, are not well established. Thus, he main objective of this research is to estimate the longitudinal changes in aggregate exposures to targeted pesticides for selected preschool children in the same age group over four years.

The variability among preschool children’s exposures in similar microenvironments and among those in different microenvironments is also not well known. Thus, a second objective of this research is to estimate the interpersonal variability of these exposures among preschool children living in different homes and among preschool children living in the same home.

Progress Summary:

The longitudinal research study, Pesticide Exposures of Preschool Children Over Time (PEPCOT), began in June 2002. In the first project year, project protocols, materials, and standard operating procedures were prepared; Institutional Review Board (IRB) approval, EPA Protection of Human Subjects approval, and an NIH Certificate of Confidentiality were obtained. Also in the first project year, participants were recruited and Year 1 field sampling was completed for the spring group of participants.

Recruitment

Fifty households, as planned, were recruited into the study. Two or more children per household are participating in PEPCOT, including six sets of twins and one set of triplets, for a total of 101 children. In the second project ear, IRB approval for continuation of the project was obtained. Sampling appointments were scheduled for the Year 2 summer group. Two participants dropped out of Year 2 field sampling, one each from the spring and summer sampling groups, for a drop-out rate of 4 percent. Currently, 48 households remain in the study.

Field Sampling

The 50 participating households were each assigned to one of three groups for spring, summer, or fall sampling. By the end of May 2004, Year 1 field sample collection was completed for all households in all three groups, and Year 2 sampling was completed for 16 households in the spring group of participants. Samples were collected over a 24-h period. Microenvironmental samples included: indoor and outdoor air, indoor floor dust, food-preparation surface wipes and floor surface wipes. Personal samples included children’s liquid and solid food, hand wipes, and first morning urine. For those children still in diapers, overnight diapers were collected. For those children being breast-fed, samples of breast milk were requested, although not always possible to obtain.

Additional information collected includes child activity diaries, observations of house/building characteristics, and information on household, family, and residence characteristics and activities that may affect the children’s exposures, plus brief information on the children’s health that may be relevant to exposure measurements.

Laboratory Chemical Analysis

Chemical analysis of the collected samples is in progress. Analysis was completed for all Year 1 samples of indoor and outdoor air, urine except diapers, and hand wipes. These data are being imported into the PEPCOT database

Future Activities:

In the third year of this project, Year 2 field sampling will be completed with the summer and fall groups of households, and Year 3 sampling will be initiated with the spring group of households. Laboratory analysis of the collected samples, analysis of the data, and manuscript preparation for publication of PEPCOT results will continue.
