Objective:
The objective of this research project is to elucidate the role of nonprotein, biologically relevant manganese(II)/(III) complexes in DNA damage and/or protection when exposed to: (1) an intracellular oxidant (hydrogen peroxide); and (2) an environmentally available oxidant system (sulfite/oxygen).
Approach:
Manganese(II)-DNA reactivity in the presence of hydrogen peroxide and carbonate, in phosphate or pyrophosphate buffer, was examined by polyacrylamide gel electrophoresis (PAGE). Manganese(II)-DNA reactivity in the presence of sulfite also was examined by PAGE. Radicals generated in the system without DNA were detected by electron spin resonance (ESR) trapping experiments. To identify the DNA moiety at which the reaction occurs, the reaction of DNA monomers was investigated by ESR spin trapping, 31P nuclear magnetic resonance, and electrospray mass spectrometry.
