Objective:
WORK COMPLETED (June 1 to Aug. 31, 2001)
- The Smart Cycler was delivered and installed.
- The performance of the Smart Cycler has been studied with appropriate standards to establish its reliability.
- Two sets of primers have been secured and tested.
- Extraction and analysis of ribosomal DNA have been conducted on several microbial strains.
- Evaluation of the Smart Cycler in our laboratory has shown that it is fully capable of quantifying the level of dioxygenase enzymes present in micobial isolates and soil samples taken from contaminated field sites.
- Preliminary runs with primers have provided results that will be used to redesign the primers for more effective quantification of dioxygenase genes.
RESULTS
SIGNIFICANCE
Quantitative PCR determinations of ribosomal and nah genes have been made for the first time on plant rhizosphere samples.
