Objective:

Objective Project 1 (Environmental Epidemiology)

To build upon our discovery of immunologic and molecular biomarkers specific to children with autism found in 2-5 year olds enrolled in the CHARGE (Childhood Autism Risks from Genetics and Environment) Study.

Objective Project 2 (Immunological Susceptibility of Asthma)

Our current aims include the continued examination of environmental toxicants such as PBDE47 on immune function in children with autism. In addition, we are currently working on the identification of the autoantigens in both the maternal population and their offspring. We will bring back our subjects for a longitudinal analysis of their immune function parameters studied previously. We have been performing B cell function analysis using PBMC from CHARGE-BACK subjects. We are currently waiting to obtain more typically developing (TD) subjects for this aspect of the study prior to data analysis. 

Objective Project 3 (Neurodevelopmental Toxicology)

The specific aims of Project 3 remain essentially unchanged. We have added a new investigator, Professor Pamela Lein, who will be looking at the interactions between organophosphorus pesticides (OPs) and neuroligins. The overall goal is to determine if exposure to environmental toxicants early in development contributes to the etiology of neurodevelopmental disorders such as autism. A related goal is to determine whether susceptibility to autoimmune disease increases the neurotoxicity of environmental contaminants and increases the risk for development of neurodevelopmental disorders. Specific Aim 1 examines perinatal exposure to BDE47, PCB 95 and mercury in C57BL/6J and SJL/J mice for effects on spatial memory and learning, sensory and motor development, sensory gating and fear/anxiety. Specific Aim 2 examines whether neonatal exposure to environmental contaminants alters immune system function, determined by exposing mice to toxins and measuring immunoglobulins, cytokines and chemokines. Specific Aim 3 examines whether neonatal toxin exposure alters brain excitability. Specific Aim 4 is designed to determine whether injection of immunoglobulins isolated from the serum of mothers of autistic children into pregnant mice alters brain development and behavior. A decision has been made to drop the SJL/J mouse strain from these studies, and to focus on the neurodevelopmental effects of perinatal toxin exposure in C57BL/6J mice. This decision was based on concerns that blindness by 4 weeks of age in the SJL/J mouse strain, which carries the retinal degeneration allele Pde6brdl, presented an unacceptable limitation to the use of the SJL/J mouse in neurobehavioral studies. This decision was discussed with the external advisory committee for the CCEH, which also advised this change in the proposed experiments. 

 

Progress Summary:

Project 1

Specific Aim 1. Hypothesis:  Children with autism can be distinguished from those without autism by markers of immune dysregulation (at birth, as well as post-diagnosis) and by prenatal, immunologically relevant events and exposures.

Substantial work was conducted during year 10 on this aim. Using data from the CHARGE Study, we have completed work on a number of immune markers comparing children with and without autism. In vitro stimulation of peripheral blood mononuclear cells collected from confirmed cases and controls resulted in differential production of GM-CSF, TNF-alpha, and IL-13, all of which were increased in specimens from children with ASD as compared with those from typically developing controls (Ashwood et al., Brain Behav Immun 2010). Moreover, pro-inflammatory cytokines were associated with worse behaviors in core domains for ASD. In contrast, GM-CSF and TH-2 cytokines were associated with improved cognitive function. Additionally, several chemokines (MCP-1, RANTES, eotaxin) were elevated in plasma of affected children and associated with deviant behaviors and poorer developmental scores (Ashwood et al., J Neuroimmunology 2011). Moreover, using Western blot, we found that children with autism or ASD produce distinct patterns of plasma auto-antibodies to cerebellum proteins and that the presence of these antibodies correlates with worse behaviors and lower scores in cognitive and adaptive domains (Goines et al 2011, Autoantibodies to cerebellum in children associate with behavior).

Taking an epidemiologic approach, we have examined several prenatal immunologically relevant events in relation to autism. First, using statewide data on over 6 million births, we showed that the date of conception for children who developed autism was more likely to be in the winter months, December through March (Zerbo et al., Epidemiology 2011). This finding is consistent with an early infectious exposure. Second, mothers of CHARGE Study children with confirmed autism were more likely to report having had flu in the second trimester (though this did not reach statistical significance). They were also more likely to report fever, but only if the fever was not treated with medication:  in children of those with fever who did medicate with antipyretics, the risk of autism was no different from offspring of mothers without fever, but in children whose mothers had a fever but did not medicate, the odds ratio was over two-fold (Zerbo et al. in submission).

Third, we examined several maternal chronic conditions that are associated with inflammation:  diabetes or gestational diabetes, hypertension, and obesity. Children with autism were more likely to have a mother who had one or more of these conditions, with an OR of 1.61, 95% CI=(1.10, 2.37). These conditions were also associated with greater risk for developmental delay and with lower scores in expressive or receptive language (Krakowiak et al, in review).  

This project also collaborated with Project 2 in collection and analysis of specimens for the CHARGE-Back study, which examined continuity of markers of dysregulated immune function. 

Specific Aim 2. Hypothesis:  Children with autism can be distinguished from children without autism by their xenobiotic exposures and biomarkers, which in turn are related to immune dysregulation.

Over the last year, we have continued our investigations of mercury, air pollution and nutritional factors. We completed analyses of baby hair specimens and validated their correlation with fish consumption. Some difficulties with the solid standards became apparent with the second batch of hair specimens that were analyzed, and Dr. Green has had to develop our own standards for this work. That development is complete, and the re-analysis of batch two along with a third batch will complete these analyses. To date, using the high-quality batch 1 set, we saw no association of infant hair mercury levels with diagnostic group (autism, developmental delay, typical development) (McKean et al., in preparation). A second study used newborn blood spots and also found no association with autism risk, and no evidence of an interaction with child’s genotype for GSTM1 (McKean et al., in preparation). We are expanding our sample size, based on the additional shipment of more than 650 more newborn bloodspots. A third manuscript has investigated the use of the bloodspots and the questionnaires to develop a toxicokinetic model for fish levels of mercury in pregnant women (McKean et al., in preparation).

Air pollution and proximity to traffic as a proxy for exposure to motor vehicle emissions were also examined using the CHARGE Study participant addresses during pregnancy and at the time of delivery. Mapping of addresses and geocoding to calculate distance to the nearest freeway revealed that case families were nearly twice as likely to reside within 300 m of a freeway as compared with control families (OR = 1.86, 95% CI = 1.04, 3.45) (Volk et al., Env Health Persp 2011). Further work has been conducted using estimated exposures to fine particles using several exposure databases and results indicate similar associations (Volk et al., in review).

With regard to household products, an in-depth analysis of pesticides demonstrates a consistent association between household pyrethroid-containing insecticides during the gestational period and risk for autism. The risks exceeded three-fold for those who used these products regularly (e.g., 3 months in the first and second trimester, or 6 months in the full pregnancy). We found that associations were weaker when pyrethrin-only insecticides were evaluated, but stronger when restricted to those who were born 2002 or later (after the banning of organophosphates for household products) or in those boys who carried the 4-repeat allele in the promoter region of the MAOA gene. These differentials in the associations suggest that recall bias is unlikely to explain the findings.  

Finally, maternal report of prenatal vitamin supplementation was assessed in relation to risk for autism or ASD. Results showed that mothers of affected children were less likely to report having taken prenatal vitamins in the period from three months prior to conception through the first month of pregnancy (Schmidt et al., 2011). By the end of the first trimester, the vast majority of mothers of both cases and controls were taking these vitamins. Thus, this finding points to a very early window of susceptibility. Also, if either the mother or the child inherited a genotype for one of the critical genes encoding enzymes in the one-carbon-metabolism cycle that involved less efficient transfer of methyl groups to DNA, the child was at much higher risk when the mother had not supplemented with vitamins in the critical window (Schmidt et al., 2011).

Specific Aim 3. Hypothesis:  Transcriptional genomic profiles of children with autism differ from those of children without autism.

Transcriptional analysis of CHARGE Study samples was reported in a previous year, and  several manuscripts were published suggesting that the relationship of gene expression to mercury and lead differs by case status (Tian et al., 2011, Stamova et al., 2011). Similar analysis on MARBLES subjects will await further information on final diagnoses, in order to maximize our statistical efficiency by choosing an efficient ratio of cases: controls.

Specific Aim 4. Hypothesis:  The exposure and biomarker (immune, xenobiotic, genomic) differences will be most pronounced for specific phenotypic subsets among children in the CHARGE study.

Significance:

In the last year, analysis of the CHARGE Study specimens and database has provided convincing evidence regarding immunologic dysregulation leading to increased pro-inflammatory activity in children with autism, and a statewide analysis of children receiving services for autism suggests a seasonal contribution to increased risk based on conception in the winter months. Additional analyses of the CHARGE Study indicate that second trimester fever, uncontrolled by medication, may also confer higher risk of autism in the offspring. Similarly, chronic inflammatory conditions of the pregnant mother may predispose the child to a higher risk for ASD. These findings together support a potential inflammatory involvement potentially originating in the prenatal period, and either leading to or exacerbating neuropathologic events.

Our findings with regard to xenobiotic chemical and particle exposures may provide opportunities for interventions. First, traffic and estimated air pollution concentrations appear to be associated with elevated risk for ASD, suggesting that our current regulations may not be sufficient to protect the developing fetus. Household pyrethroid insecticides, synthesized for high toxicity mediated by voltage-sensitive sodium channels and also known for interference with GABA-receptors, were more commonly used by families whose child developed autism than by those whose child showed typical development. These compounds are currently advertised as being safe and natural. The particularly high risk associated with frequent use and in those with a high risk genotype are of concern. In contrast, maternal consumption of prenatal supplements appears to have a protective influence on child development, with reduced risk of autism when these were taken in the periconception. GeneXenvironment interactions provide further evidence that the findings are not due to differential reporting by parents according to case-control status of the child. The potential for intervention on these factors as an avenue to decrease incidence of ASD should be pursued.

Project 2

(1) Effects of in vitro BDE 49 exposure:  To examine the effect of PBDE 49 on immune cell function in vitro, we have expanded our studies of immunotoxicant sensitivity to BDE49 based on some data from the Pessah and Lein labs that suggest increased toxicity on cultured neurons (Kim et al., 2011). We have noted that BDE49 has greater effects on immune function, particularly in the ASD population, at much lower concentrations than BDE47. Our current studies suggest that peripheral blood mononuclear cells isolated from children with autism have an increased sensitivity to this more potent congener that manifests in a profound dysregulation of cytokine production following in vitro exposure (Figure 1). Furthermore, we have found that exposure to various high concentrations of BDE-49 interfere with calcium re-uptake into human, primarily monocyte cultures, and inhibits the release of INF-gamma from T lymphocytes in real-time. The presentation of this work won second place for predoctoral candidates at the Neurotoxicology meeting in 2011.

(2) Maternal autoantibodies to fetal brain proteins: The presence of these antibodies in the plasma of some mothers of children with autism might suggest an association between the transfer of IgG autoantibodies during early neurodevelopment and the risk of developing autism in some children. We have now identified the 37, 39 and 73 kDa proteins recognized by maternal antibodies and are in the final stages of confirmation of these antigens. We have recently published a paper describing an expansion study wherein we tested additional mothers for antibodies to fetal brain proteins. In addition, we found that those mothers with the 37/73 kDa pattern had children with greater expressive language deficits (Braunschweig et al., 2011). We now have a published patent on the identity of these antigens.
 
Also published this past year was a study describing the relationship between the MET C/C allele polymorphism and the presence of antibodies to the 37/73 kDa fetal brain antigens (Heuer, et al, 2011). This study is the first to describe a relationship between two risk factors for autism (Table 1).

(3) Cytokine production:  We have recently published a paper describing the plasma cytokine profile of subjects with autism and typically developing controls. We noted significant differences in plasma protein levels of a number of cytokines, including IL-1b, IL-6, IL-8 and IL-12p40 in the ASD group compared to TD controls (p < 0.03) (Ashwood et al., 2011). Moreover, when the ASD group was separated based on the onset of symptoms, it was noted that the increased cytokine levels were predominantly in ASD children who had undergone a period of normal development followed by regression. In addition, increasing cytokine levels were associated with more impaired social interactions and communication as well as more aberrant behaviors. This study was named among the TOP 10 Discoveries in Autism by Autism Speaks. We have also described similar findings with respect to plasma chemokine levels (Ashwood, et al., 2011).

In conclusion, we continue to define the immune profile of children with ASD as well as the maternal autoantibody profile. We have also determined a differential sensitivity to the PBDE family of persistent organic pollutants, and will continue to expand our mechanistic studies in collaboration with the Pessah lab. The maternal antibody antigen identification paper will be published in early 2012.

Project 3

Molecular and Cellular Toxicology

Polybrominated diphenyl ethers (PBDEs) are widely used flame retardants that bioaccumulate in human tissues. Their neurotoxicity involves dysregulation of calcium ion (Ca2+) signaling; however, specific mechanisms have yet to be defined. We aimed to define the structure–activity relationship (SAR) for PBDEs and their metabolites toward ryanodine receptors type 1 (RyR1) and type 2 (RyR2) and to determine whether it predicts neurotoxicity.

Methods:  We analyzed [3H]ryanodine binding, microsomal Ca2+ fluxes, cellular measurements of Ca2+ homeostasis, and neurotoxicity to define mechanisms and specificity of PBDE-mediated Ca2+ dysregulation. PBDEs possessing two ortho-bromine substituents and lacking at least one para-bromine substituent (e.g., BDE‑49) activate RyR1 and RyR2 with greater efficacy than corresponding conge­ners with two para-bromine substitutions (e.g., BDE‑47). Addition of a methoxy group in the free para position reduces the activity of parent PBDEs. The hydroxylated BDEs 6‑OH-BDE‑47 and 4´-OH-BDE‑49 are biphasic RyR modulators. Pretreatment of HEK293 cells (derived from human embryonic kidney cells) expressing either RyR1 or RyR2 with BDE‑49 (250 nM) sensitized Ca2+ flux triggered by RyR agonists, whereas BDE‑47 (250 nM) had negligible activity. The divergent activity of BDE‑49, BDE‑47, and 6-OH-BDE‑47 toward RyRs predicted neurotoxicity in cultures of cortical neurons. We found that PBDEs are potent modulators of RyR1 and RyR2. A stringent SAR at the ortho and para position determined whether a congener enhanced, inhibited, or exerted nonmonotonic actions toward RyRs. These results identify a convergent molecular target of PBDEs previously identified for noncoplanar polychlorinated biphenyls (PCBs) that predicts their cellular neurotoxicity and therefore could be a useful tool in risk assessment of PBDEs and related compounds.

Aroclor 1254 (A1254) interferes with normal dendritic growth and plasticity in the developing rodent brain, but the mechanism(s) mediating this effect have yet to be established. Non-dioxin-like (NDL) polychlorinated biphenyls (PCBs) enhance the activity of ryanodine receptor (RyR) Ca2+ channels, which play a central role in regulating the spatiotemporal dynamics of intracellular Ca2+ signals. Ca2+ signaling is a predominant factor in shaping dendritic arbors but whether potentiation of RyR activity by PCBs influences dendritic growth is not known. We determine whether PCB effects on dendritic growth and RyRs are causally related. Using Golgi analysis of hippocampi from weanling rats we confirmed that developmental exposure via the maternal diet to NDL PCB 95, a potent RyR potentiator, phenocopies the dendrite promoting effects of A1254 in pyramidal neurons. Dendritic growth in dissociated cultures of primary hippocampal neurons and in hippocampal slice cultures is similarly enhanced by PCB 95 but not by PCB 66, a congener with negligible effects on RyR activity. The dendrite promoting effects of PCB95 are evident at concentrations as low as 2pM and are completely prevented by either blocking RyR channels with FLA365 or by siRNA knockdown of RyR expression. These findings demonstrate that environmentally relevant levels of NDL PCBs modulate neuronal connectivity via RyR-dependent effects on dendritic arborization and identify RyR channel dysregulation as a novel mechanism contributing to dysmorphic dendritogenesis associated with heritable and environmentally triggered neurodevelopmental disorders.  

In a related study, we discovered that non-dioxin-like (NDL) polychlorinated biphenyls (PCBs) promote dendritic growth in hippocampal neurons via ryanodine receptor (RyR)-dependent mechanisms; however, downstream signaling events that link enhanced RyR activity to dendritic growth are unknown. Activity-dependent dendritic growth, which is a critical determinant of neuronal connectivity in the developing brain, is mediated by Ca2+-dependent activation of Ca2+/calmodulin kinase-I (CaMKI), which triggers cAMP response element binding protein (CREB)-dependent Wnt2 transcription. RyRs regulate the spatiotemporal dynamics of intracellular Ca2+ signals, but whether RyRs promote dendritic growth via modulation of this signaling pathway is not known. We further tested the hypothesis that the CaMKI-CREB-Wnt2 signaling pathway couples NDL PCB-enhanced RyR activity to dendritic arborization. Ca2+ imaging of dissociated cultures of primary hippocampal neurons indicates that PCB 95, a potent RyR potentiator, enhances synchronized Ca2+ oscillations in somata and dendrites. As determined by western blotting and qPCR, PCB 95 also activates CREB and upregulates Wnt2. Blocking CaMKK, CaMKIα/g, MEK/ERK, CREB or Wnt2 prevents PCB 95-induced dendritic growth. Antagonism of γ-aminobutyric acid (GABA) receptors with bicuculline phenocopies the dendrite-promoting effects of PCB 95, and pharmacological antagonism or siRNA knockdown of RyR blocks bicuculline-induced dendritic growth in dissociated and slice cultures of hippocampal neurons. These findings demonstrate that RyR activity contributes to dynamic remodeling of dendritic architecture in response to NDL PCBs via CaMKI-CREB-Wnt2 signaling, and identify PCBs as candidate environmental risk factors for neurodevelopmental disorders, especially in children with heritable deficits in calcium signaling associated with autism.

Behavioral Toxicology

Genetic factors have been established as contributing to the development of autism. We examined three loci, serotonin transporter (SLC6A4), dopamine b-hydroxylase (DBH), and the variable number of tandem repeat promoter of the monoamine oxidase A (MAOA) for association with autism in participants from the Childhood Autism Risks from Genetics and the Environment (CHARGE) Study, the first large-scale population-based case-control investigation of both environmental and genetic contributions to autism risk. Among male children enrolled in the CHARGE study we tested associations between each of the three polymorphisms and autism (AU) (n5119), or a combined group of autism and other autism spectrum disorders (AU1ASD, which includes an additional n553) as compared with typically developing controls (TD, n5137). The case-control association analysis showed neither SLC6A4 nor DBH to be statistically significantly associated with AU or ASD. However, the male children carrying 4 tandem repeats in the promoter region of the MAOA gene showed a two-fold higher risk of AU (or AU1ASD) than those carrying allele 3, adjusted for confounders (OR52.02, 95% CI51.12, 3.65, P50.02 for AU vs. TD, and OR52.05, 95% CI51.19, 3.53, P50.01 for ASD vs. TD). In addition, mothers homozygous for the 4 tandem repeat allele showed at least a three-fold higher risk of AU (or AU1ASD) than mothers homozygous for allele 3 (OR53.07, 95% CI51.19, 7.91, P50.02 for AU vs. TD, and OR53.26, 95% CI51.35, 7.89, P50.009 for AU1ASD vs. TD). These results suggest a potential role of the functional MAOA promoter alleles in the male child, the mother, or both in ASD.

Neurodevelopmental toxicity of perinatal exposure to BDE47 was examined in a series of behavioral, histological and analytical studies in mice (Ta, et al., 2011). C57BL/6j mice were exposed to 0.03, 0.1 or 1.0 mg/kg/day BDE47 (p.o.), or vehicle control throughout gestation and lactation, and the offspring examined for behavioral or neurohistological abnormalities. Perinatally exposed offspring had substantial accumulations of BDE47 in blood and brain when examined on postnatal days (PNDs) 1, 7, 14 & 21, with brain levels equaling or exceeding that of their dams at the time of birth (366 ± 117 versus 306.6 ± 60 ng/g tissue, respectively). We also found evidence for mobilization of BDE47 in the milk of lactating dams, further documenting the risk of exposure to BDE47 through lactation. Finally, offspring showed abnormal locomotor activity in the open field as well as altered performance in the Morris water maze. This study was followed up by a second study (Koenig, et al. NBTS abstract, manuscript in preparation) using the same dosing scheme for BDE47, but examining tissue levels, motor performance and learning impairments in greater detail. This second study showed high tissue levels of BDE47 in offspring, even at embryonic day 15. High levels of BDE47 were found in the blood, brain and fat of dams and offspring. Clear evidence for mobilization of BDE47 into milk during lactation was found in this study, with tissue concentrations falling dramatically in all tissue compartments during lactation, while levels in milk remained elevated. These data show that transfer of BDE47 from mother to offspring is a significant route of exposure, and also suggest that tissue levels in lactating females may underestimate their exposure history and the amount of toxin being delivered to offspring through the milk. A third study was also completed that examined misoprostol for possible neurodevelopmental toxicity. Misoprostol is a synthetic prostaglandin that is commonly given to pregnant women to induce labor. When given early in gestation it can result in Mobius syndrome, a disorder characterized by congenital palsy of the 6th and 7th cranial nerves, and autistic like symptoms in some children. In addition, the human CHARGE study found that more mothers of children with autism were exposed to misoprostol more frequently than those in the general population. Therefore, we hypothesized that neonatal exposure to misoprostol may be a risk factor for neurodevelopmental disorders such as autism. To test this hypothesis we examined the effects of exposure to 0.4, 4 or 40 µg/kg misoprostol, or saline vehicle (s.c.) on brain development and behavior. These doses were selected to span the range of doses reported clinically during the induction of labor. The results did not indicate any neurodevelopmental toxicity in our C57BL/6j mouse model, and therefore did not support our initial hypothesis. While the results were negative we feel that they have real value for the field because they serve to direct future research efforts to other environmental and pharmacological agents that may be contributing to the etiology of neurodevelopmental disorders. 

Significance:
 
We have noted several significant findings. The first is that IgG isolated from mothers at high risk of giving birth to a child with ASD and possessing autoantibodies directed at brain proteins at 70 and 33kDa were found to influence developmental outcomes in mice when the IgG was administered to pregnant mice at mid gestation. These effects were not observed in the controls exposed to IgG from mothers at low risk and lacking the autoantibodies. With respect to our work with BDEs and PCBs, we have completed the first low-dose developmental exposure study in mice with BDE 47 and have identified behavioral impairments. Finally we have completed a series of studies defining the mechanisms by which PCBs influence ryanodine receptor at the molecular level, and how these effects influence excitability of neuronal circuitry, and how these relate to alteration in basal and activity dependent dendritic growth. 

Future Activities:

Project 1

The MARBLES project will continue with sample and data collection. For the CHARGE-Back study, we will continue to work with Project 2, Immunology, to determine whether the immune profiles of these children change over time. The analysis of specimens from the CHARGE study will continue with the other cores and projects. Project 1 will provide support for the preparation of manuscripts that address, e.g., immune and gene expression markers in CHARGE study children, comparing ASD to typically developing pre-schoolers.

Project 2

We will continue to expand our maternal autoantibody study into a separate RO1 for our work with Campbell et al and the association of these specific antibodies with the presence of the MET C polymorphism in conjunction with the MARBLES study. We will continue our PBDE49 study, which has been demonstrated to be more immunotoxic. We will begin developing an animal model using the defined fetal brain protein antigens, both with and without toxicant exposure. 

Project 3

The current period of no funding will be used to conclude the activities of this project.
