Objective:

Objective of Research Project 1: 

To build upon our discovery of immunologic and molecular biomarkers specific to children with autism found in 2-5 year olds enrolled in the CHARGE (Childhood Autism Risks from Genetics and Environment) Study.

Objective of Research Project 2: 

Our current aims include the continued examination of environmental toxicants such as PBDE47 on immune function in children with autism. In addition, we currently are working on the identification of the autoantigens in both the maternal population and their offspring. We will bring back our subjects for a longitudinal analysis of their immune function parameters studied previously. We have been performing B cell function analysis using PBMC from CHARGE-BACK subjects. We currently are waiting to obtain more typically developing (TD) subjects for this aspect of the study prior to data analysis. 

Objective of Research Project 3: 

The specific aims of Project 3 remain essentially unchanged. The overall goal is to determine if exposure to environmental toxicants early in development contributes to the etiology of neurodevelopmental disorders such as autism. A related goal is to determine whether susceptibility to autoimmune disease increases the neurotoxicity of environmental contaminants and increases the risk for development of neurodevelopmental disorders. Specific Aim 1 examines perinatal exposure to mercury, BDE47 and PCB 95 in C57BL/6J and SJL/J mice for effects on spatial memory and learning, sensory and motor development, sensory gating and fear/anxiety. Specific Aim 2 examines whether neonatal exposure to environmental contaminants alters immune system function, determined by exposing mice to toxins and measuring immunoglobulins, cytokines and chemokines. Specific Aim 3 examines whether neonatal toxin exposure alters brain excitability. Specific Aim 4 is designed to determine whether injection of immunoglobulins isolated from the serum of mothers of autistic children into pregnant mice alters brain development and behavior.

Progress Summary:

Project 1

Newborn blood spots: More than 300 of the CHARGE study newborn bloodspots have been analyzed in the laboratory of Dr. Peter Green at University of California-Davis (UC Davis) for metals, both Hg and Pb.

Blood mercury concentrations were compared across developmental groups. The comparison shows that children with autism do not differ from those without autism in their current blood Hg levels. A manuscript has been submitted.

Candidate gene studies: Trios consisting of a proband (case or control) and two parents have been genotyped for several candidate genes including GSTM1, 5-HT-receptor, dopamine hydroxylase, and MAOA. The results indicate that neither 5-HT nor dopamine hydroxylase genes are associated with autism in association analyses, but that a polymorphism of allele 4 for the MAOA gene (located on the X chromosome) shows a different frequency.

Pesticides: An analysis of household products with an emphasis on pesticides is under way. Preliminary results demonstrated that children of mothers who used pet tick/flea control shampoos or soaps during pregnancy or the first year of the child’s life had a two-fold higher risk of developing autism. These products contain high levels of pyrethrins, which, in experimental studies, have been shown to cause neuronal death and to compromise the blood brain barrier. This paper will be presented at IMFAR.

PBDEs: The data analysis phase is nearly complete on the 100 children in whom we measured brominated flame retardant concentrations in serum. The results indicate a somewhat lower concentration of the most highly brominated congeners, but no difference in other congeners. Concentrations also are lower in children with developmental delay. We also have begun to explore the relationship between brain volume (using head circumference as a marker) and levels of these highly brominated congeners.

Folate: An initial analysis of folate intake, as reported by mothers on the Environmental Exposure Questionnaire, indicates that supplementation in the 3 months prior to conception and the first month after conception confers a significant protection against autism. Supplementation at other points in pregnancy confers no protection. Further work to delineate the dosages and conduct a dose-response analysis currently is in progress.

Air pollution: In collaboration with the USC Center for Children’s Environmental Health, we are examining air pollution and autism. Traffic derived air pollutant exposure assessment will be determined by calculating distance from the nearest freeway, highway, and/or major arterial road using geo-coded address data. Exposure estimates will be calculated for autism cases and for matched control subjects. These distance measures will be associated with diagnoses of autism, developmental delay, and underlying autism-related traits (e.g., social functioning).

Fieldwork for the CHARGE-BACK study is under way. A total of 57 primary children, their parents and siblings have come to the clinic, for a total of 178 CHARGE-Back participants to date.

The MARBLES (Markers of Autism Risk in Babies – Learning Early Signs) Study, a longitudinal cohort study of pregnancies of high-risk children was launched, after a pilot phase. The pilot phase began recruitment in July 2006, supported by funding from the M.I.N.D. Institute. To date, we have enrolled nearly 70 women, conducted more than 190 home visits, had 38 deliveries and obtained specimens from 35 of them. We have completed 25 first trimester visits, 39 second trimester visits, and 51 third trimester visits, collecting urine, hair, blood, and saliva. Prenatal home dust collections have been completed for 35 subjects. Postnatal specimen collections include 35 2-week baby visits, 24 3-month visits, 13 6-month visits, and 4 one-year visits. At these visits, we have successfully collected baby and maternal blood, urine, saliva, and hair, as well as conducted baby assessments. We also have completed eight additional post-natal dust samples. Additionally, hundreds of maternal weekly symptom diaries, and dozens of monthly baby diaries, have been completed.

We have received support from the UC Davis General Clinical Research Center (GCRC) in the Clinical Translational Science Center, which now provides staff to attend home visits and carry out maternal and pediatric phlebotomy services for our study subjects. Procedures related to environmental sampling have been refined and dust samples have been collected at home visits.

An application to the National Institutes of Health (NIH) for a multi-site study modeled after the MARBLES Study was funded as an Autism Center of Excellence (ACE) Network. This project, “Early Autism Risk Longitudinal Investigation” or EARLI, began in 2008 (1 R01-HD055803), with Dr. Craig Newschaefer at Drexel University as PI. UC Davis will be one of four sites and will house the Data Coordinating Center.

Analysis of CHARGE samples is continuing, and the newborn bloodspot analysis will provide the first data ever on prenatal Hg exposures for individual children with and without autism (bloodspot study). If the results on folate intake turn out to be robust, identification of a modifiable protective factor for autism may be highly significant. Similarly, the CHARGE Study provides the first evidence to date that mothers of confirmed autism cases are more likely to report having used certain pest control products more frequently than mothers of typically developing controls. The growth in use of pyrethrin pesticides is beginning to manifest as widespread home contamination, and deserves serious attention for the potent toxicities of these compounds. The first longitudinal study of autism with environmental and molecular factors measured in specimens collected in pregnancy (MARBLES) is under way and immunologic studies in mothers and the offspring have begun. Determination of whether immunologic differences between children with autism and other children are persistent (CHARGE-BACK) will shed light on whether immune dysfunction is incidental or integral to this complex disorder.

Project 2

The aim is to examine the effect of in vitro xenobiotic exposure such as PBDE47 on peripheral blood mononuclear cell (PBMC) function. We have noted a change in the response of PBMCs from subjects with autism compared to controls when in the presence of PBDE47. There is a profound change in the response to LPS by the monocyte/macrophage population in the autistic children (AU) population compared to the typically developing children (TD) controls. This suggests that they are extremely sensitive to such exposure and this could ultimately affect immune function.

The presence of these antibodies in the plasma of some mothers of children with autism, as well as the differential findings between mothers of children with early onset and regressive autism may suggest an association between the transfer of IgG autoantibodies during early neurodevelopment and the risk of developing autism in some children.

We currently are using a proteomic approach to begin antigen identification. We have taken advantage of a new protein separation device from Bio-Rad to size-fractionate our crude monkey brain for better antigen preparation. This preparation has been used in an affinity mass spectrometric approach to perform peptide mapping. The figure below illustrates the protein separation that was used for proteomic analysis. We have focused first on the 37 kDa antigen based on its high specificity. We also have discovered a new antigen at 39 kDa associated with maternal antibodies and an early onset diagnosis in the child. Finally, in a collaborative effort with Drs. Pat Levitt and Dan Campbell at Vanderbilt University, we have preliminary data suggesting a strong association between the MET C allele polymorphism and the presence of maternal antibodies to the 37 kDa fetal brain protein antigen (genotypic c2=7.166; df=2; P=0.028; allelic c2=8.420; df=1; P=0.004). We currently are extending this study and seeking additional funding in a new RO1.

We have noted reduced levels of plasma IgG and IgM in the AU population compared to both DD and TD controls suggestive of an underlying defect in immune function. This reduction in specific Ig levels correlates with behavioral severity, where those patients with the highest scores in the ABC behavioral battery have the most reduced levels of IgG and IgM. Plasma from children diagnosed with autism, ASD, developmental delay, or typically developing controls, were assayed for levels of total immunoglobulin by ELISA. Children with autism displayed a significantly reduced level of total IgG when compared to both typically developing controls and children with developmental delay. Total IgM levels also were significantly reduced in children with autism compared to typically developing controls. In contrast, both total IgA and IgE levels showed no difference between any of the groups. This manuscript currently is out for review at Autism Research. We now are looking at the B cell population following stimulation with various B cell specific mitogens. In our very preliminary data, we have noted a decrease in the CD19+ B cell population following stimulation. We currently are expanding this study.

We examined plasma from children with ASD, as well as age-matched typically developing controls, for antibodies directed against human cerebellar protein extracts using western blot analysis. In addition, the presence of cerebellar specific antibodies was assessed by immunohistochemical staining of sections from Macaca fascicularis monkey cerebellum. Western blot analysis revealed that 13/63 (21%) of subjects with ASD possessed antibodies that demonstrated specific reactivity to a cerebellar protein with an apparent molecular weight of approximately 52kD compared with only 1/63 (2%) of the typically developing controls (p=0.0010). Intense immunoreactivity, to what was determined morphologically to be the Golgi cell of the cerebellum, was noted for 7/34 (21%) of subjects with ASD, compared with 0/23 of the typically developing controls. Furthermore, there was a strong association between the presence of antibodies reactive to the 52 kDa protein by western blot with positive immunohistochemical staining of cerebellar Golgi cells in the ASD group (r= 0.76; p=0.001) but not controls. These studies suggest that when compared with age-matched typically developing controls, children with ASD exhibit a differential antibody response to specific cells located in the cerebellum and this response may be associated with a protein of approximately 52 kDa. This paper was recently accepted in Brain Behavior and Immunity.

We have noted several significant findings during the past year. The first is the presence of maternal autoantibodies in a subset of subjects that potentially could be used as a diagnostic marker for autism risk. The second is the relationship between behavior and immune dysfunction as noted by the correlation between IgG and ABC score. This suggests to us that there is a strong link between immune dysfunction and severity of autism. We also have noted that staining of Golgi cells in the cerebellum corresponds to the presence of a 52 kDa band in cerebellar extract as noted by western blot analysis. Our most recent findings suggest that in other regions of the brain, the reactive cells appear to be confined to GABAergic interneurons as determined by GAD co-localization studies. Finally, we have preliminary studies that suggest the individuals with autism are differentially affected by PBDE 47 exposure with respect to direct effects on macrophage/monocyte function.

Project 3

Effects of neonatal PBDE47, PCB95 and MeHg exposure on brain development and behavior in C57BL/6J and SJL/J mice. We have completed a large and comprehensive examination of the effect of neonatal exposure to the polybrominated diethylether flame retardant (PBDE47) on brain development and behavior in C57BL/6J mice. A manuscript is in preparation detailing these studies, and a brief summary of the results is presented below. Polybrominated diphenyl ethers (PBDEs) are widely used flame retardants that have become pervasive environmental contaminants. Chemical stability and lipophilicity have contributed to the bioaccumulation of PBDEs with increasing levels detected in humans and in wildlife that may represent a major health risk. Therefore, we evaluated the potential developmental neurotoxicity of PBDE47, one of the most abundant PBDE congeners identified in animal and human tissues. Female C57BL/6J mice were exposed to a daily oral dose of 0, 0.03, 0.1 or 1 mg/kg beginning 4 weeks prior to conception, continuing through gestation and lactation, and ending at weaning on PND 21. The levels of PBDE47 in blood, brain, liver and adipose tissues of dams were markedly increased after 4 weeks of exposure, around the time of mating, and continued to increase after 8 weeks of exposure, approximately the time of parturition. The levels of PBDE47 between parturition and the end of lactation, however, appeared to decrease, possibly reflecting mobilization of PBDE47 from fat stores into milk during lactation. Brain PBDE47 levels in the offspring at PND 1 approached those of the dams after 8 weeks of exposure. Perinatal exposure to PBDE47 resulted in a significant dose-dependent growth retardation (Figure 1) and an impairment in motor performance shown as a slower swim speed in the Morris water maze (Figure 2). These results indicate that developmental exposure to chronic low levels of PBDE47 resulted in significant bioaccumulation in target organs, including brain, and is associated with early deficits in exposed neonates.

There were no significant effects of PBDE47 exposure on brain development, assessed by a morphological examination of brains of mice, as well as stereological estimates of the volume and numbers of pyramidal neurons and dentate granule neurons in the hippocampus.

Effects of maternal IgG injections. Dr. Van de Water, a project investigator, had demonstrated previously that antibodies found in the blood of mothers of autistic children are reactive against fetal, but not adult, brain tissue (Braunschweig, et al., 2008). Further these antibodies are not found in the serum of control mothers (no autistic children; i.e., TD IgG). Studies were undertaken during the past year to test the hypothesis that antibodies isolated from the blood of mothers of autistic children can influence neurobehavioral development, and in particular, social behavior. To test the hypothesis, the antibodies were injected into pregnant mice and we are examining their neurobehavioral development and social interaction at the juvenile (childhood) stage of development. The brains of these animals also are being examined histologically for evidence of morphological abnormalities and for changes in cell numbers in specific brain regions (e.g., hippocampus).

In pilot studies, it was determined that multiple injections of human antibodies into pregnant mice had adverse effects on completion of pregnancy and offspring survival. Of 43 mice injected by tail vein with 1.5 µg IgG protein/g mouse body weight or vehicle on gestation day 12 and/or 16, 20 did not produce viable litters. Some indications of abnormal development were seen in the offspring of dams injected with antibodies from autistic mothers, namely larger litter size, larger individual pup size (weight, length and biparietal diameter), shorter duration of ultrasound vocalization during separation from the nest, and lower social novelty preference. However, high mortality rates in this pilot work precluded statistical comparisons. Further pilot data indicated that marginal influences on pregnancy were elicited by a single antibody injection on gestation day 12, an age implicated in environmental influences on human autism (Ingram, et al., 2005). Thus, this treatment was adopted for the full study which is ongoing.

Initial results from our pilot studies with a single antibody injection on gestation day 12 indicate that biparietal diameter (a measure of head width) in offspring of mice injected with antibodies from mothers of autistic children (AU) was significantly greater on postnatal day (PND) 28, but not on PND 14 or 18 compared to offspring of mice injected with IgG from mothers that have given birth to typically developing children (TD) and lack IgG directed at fetal brain protein (Table 1, below). This is of particular interest because macrocephaly is known to be associated with autism (Lainhart, et al., 2008).

Data from the same pilot study of mice injected once on PND 12 with antibodies from mothers of autistic children (AU IgG), or with saline or antibodies from mothers with no autistic children (TD IgG) are presented in Table 2. Pups were separated from their nest for a 2-minute period for recording of ultrasonic vocalizations. Social novelty preference was recorded during a 10-minute session at 24 days of age.

On the basis of pilot work, a full study was undertaken in January 2008. The design will compare offspring of mice injected with AU IgG antibodies to those injected with antibodies from mothers in the typically developing population (TD IgG) (n = 15/group). Each group will include antibodies isolated from three different AU mothers and three different TD mothers. In addition a saline injected control is included. The test battery includes a daily evaluation of neurobehavioral development from day 8 to 21 postnatal. After weaning from the mother on PND 21, three tests of social behavior are conducted:

1) Sociability and social novelty preference: preference for spending time in a chamber with a conspecific versus an empty chamber, or with a familiar vs. novel conspecific (Moy, et al., 2007).

2) Social dyadic interaction: interaction with another mouse of the same age scored using ethogram for juvenile behavior (Laviola, et al., 1994).

3) Social conditioned place preference: preference for a cage in which the mouse was housed with littermates versus a cage in which it was housed alone. The cages differ in the type of litter and enrichment objects present (Panksepp and Lahvis, 2000).

To date 50 mice have been injected and entered into the study, of which 9 have failed to deliver viable litters. Testing of the offspring is scheduled to be completed August 9, 2008.

Molecular mechanism and neurobehavioral consequences of developmental PCB95 exposure in mice. PCB95 is a noncoplanar PCB that can sensitize ryanodine receptors (RyR) to agents that alter intracellular calcium release. We have published evidence that auditory cortex development and excitatory-inhibitory synaptic balance are affected in offspring of rat dams fed 6 mg PCB95/kg/d (Kenet, et al., 2007). We have elucidated the molecular consequences by which ryanodine receptor type 1 (RyR1) influences the gating of individual RyR1 channels and have identified the relationship between this functional modification and the associated 3D structures of frozen hydrated RyR1-FKBP12 complexes in the open and closed states at ~9 Å resolution by cryo-electron microscopy (Samso, et al., submitted). PCB95 stabilizes the full open state of the channel by inverting the thermodynamic stability of RyR1’s closed and open states (Figure 3 a&b) permitting detailed analysis of its ion gating mechanism.

We found that the previously identified inner helices and the "inner branches" of RyR1 form a helical bundle each connected to the ion gate. Open/closed structural comparison shows that both of these structures team up to create an efficient novel mechanism of gating that consists of wringing and relaxing their two helical bundles along their common fourfold axis (see Figure 4a-f).

We have performed a broader characterization of developmental neurobehavioral effects of PCB95 in mice. In a paper we will be presenting at the Neurotoxicology and Teratology meeting later this year (Ta, et al., 2008), C57Bl/6J mouse dams (n = 7) were treated orally with 6 mg PCB95/kg/d from GD 5 to PND 21. Controls (n = 7) received corn oil. Maternal and offspring mortality and weight gain were not influenced by PCB95 exposure. Offspring (1 male and 1 female per litter) were evaluated from PND 45 to 100 with a behavioral test battery. Compared to controls, PCB95 exposed offspring had somewhat higher arousal and activity levels and showed enhanced Morris maze learning. Tests of anxiety and social interaction were not affected. These data are consistent with the earlier suggestion of altered cortical excitatory-inhibitory balance after developmental exposure to PCB95.

We have completed the first study of low-level perinatal exposure to low (relevant levels) of PBDE47. In addition to measuring its distribution to the brain, we have identified several associated behavioral deficits. We have made significant progress in transferring previous CCEH findings of developmental effect of AU IgG in the monkey (Martin, et al., 2008) to the mouse. We have found that the mouse in fact expresses the brain proteins that are the target of maternal AU IgG and are proceeding with assessing the behavioral consequences of AU IgG in the mouse. This is highly significant in that we are likely to develop a mouse AU IgG model of autism susceptibility. We have identified the molecular mechanism by which PCB95 interacts with RyR1 channels to influence both its gating function and structural conformation at 9A resolution.

The project received a supplemental award from NIEHS for $69,820 to study perinatal misoprostol exposure and increased risk for autism. The award period is August 1, 2007-July 31, 2008.

Future Activities:

Future Activities Project 1:

Recruitment for the MARBLES study will continue, and protocols will be finalized for the 24-month visits and implemented. CHARGE-BACK recruitment will continue. Manuscripts will be finalized on PBDEs, pesticides, and folate. Statistical analyses of the blood-spot data will be conducted and a manuscript prepared.

Future Activities Project 2:

We will continue to determine the maternal antibody target proteins and expand this study into a separate RO1. We also will expand our current PBDE47 findings to increase our data points and determine the correlation between these data and those found in the mouse exposure studies with Project 3. We currently are working on the identification of the maternal antibody target proteins in the hope of developing a screening assay. We will extend our findings with the MET C polymorphism into a new RO1. In addition, we also will extend the autoantibody findings in the children into a new RO1. Finally, we will continue to examine the cellular mechanism responsible for the low IgG and IgM production and determine the relationship between Ig production and behavior.
