Objective:

Aim 1: Assess the impact of in utero and peripubertal exposures to mixtures of EDCs (bisphenol A, phthalates, lead and cadmium) on linear growth and weight status (BMI); sexual maturation and reproductive hormone dynamics.

Aim 2: Determine whether EDC mixtures (BPA, phthalates, lead, cadmium) via epigenetic mechanisms induce oxidative stress (tyrosine oxidation products), disrupt metabolic homeostasis (free fatty acids, amino acids, Acyl-carnitine) and lead to changes in gene transcription and metabolic function.

Aim 3: Conduct tissue- and age-specific unbiased epigenomic analyses (DNA methylation, chromatin structure, transcriptomics) to identify a subset of tissue-independent labile genes to serve as biomarkers of exposure-induced metabolic syndrome.

Aim 4: Examine the role of dietary intake during pregnancy and adolescence in modifying the impact of EDC mixtures on metabolic homeostasis, oxidative stress and risk of metabolic syndrome.

Aim 5: Foster career development by assisting new investigators in the advancement of their research skills and knowledge in translational and children's environmental health research.

Aim 6: Share current research findings on the role that environmental exposures have on children's health in an accurate, relevant way that allows community members, healthcare professionals, and policy-makers to incorporate this new knowledge into practice for the protection of children's _health.

Progress Summary:

Research Projects: Research Projects 1 and 2 examine relationships between toxicant mixtures exposures in utero and peripuberty with physical growth, maturation and metabolic outcomes. The Early Life Exposures in Mexico to ENvironmental Toxicants (ELEMENT) cohorts compromise the primary source of human participants for Projects 1 and 2, building upon work funded through our P20 Formative CEHC. In Year 1 we established subcontracts with Yale School of Public Health and the Instituto Nacional de Salud Pública (INSP) as well as protocols for conducting fieldwork in Mexico City in our research facility at ABC Hospital, including collection of biospecimens, survey data anthropometry and measurement of maturational tempo, and completed relevant amendments to protection of human subjects. The Michigan Mother Infant Pairs (MMIP) cohort is a new US study population established by MPI Padmanabhan that is included in Project 2 of the UM CEHC. In Year 1, we prepared MMIP specimens including isolating RNA and DNA from maternal 1st trimester, maternal term, and infant cord blood samples to pursue DNA methylation and gene transcription studies and will be completing analysis of maternal urine for phthalates at 2 time points (n=80) in Quarter 4.

Project 3 is being implemented in the Agouti mouse model. In Year 1, we have established six cohorts of mating pairs and litters for our mouse model. Mice are being exposed to one of six diets to assess whether in utero high-fat diets modify the effects of perinatal BPA exposure on metabolic, hormonal, and oxidative stress parameters throughout the life-course and to identify if DNA methylation alterations result from these different diets. Additional detail is provided in the Project 3 description.

 

The Data Management and Modeling Core (DMMC) provides ongoing biostatistical support and database management for the Center to enhance investigators’ and trainees’ study designs and data analysis plans and provide technical consultation in statistical programming, presentation of methods and interpretation of findings for manuscript and poster/presentations at scientific meetings. Additional issues of common interest are discussed at Center-wide meetings.

The Community Outreach and Translation Core (COTC) held bi-monthly meetings with our partners: Head Start for Kent County, Healthy Homes Coalition, Kent County Health Department, and the Children’s Environmental Health Network. These organizations, through educational programs, work to reduce environmental health hazards for children and our partners have indicated an interest in learning about the scientific findings of the U-M CEHC to help bolster their work. As agreed in discussions with our Project Officer, Dr. Karen Peterson as Contact PI is designated as the MPI with primarily responsibility for communicating to key external stakeholders on behalf of the U-M CEHC.

Training and Professional Development: In Year 1, the University of Michigan Children’s Environmental Health and Disease Prevention Center (U-M CEHC) has provided training opportunities for undergraduate, masters, and pre-doctoral trainees to receive mentorship from senior and junior faculty members to enhance their research skills and knowledge in basic and translational research. Students are involved with laboratory work, data analysis, and manuscript preparation. By attending monthly U-M CEHC meetings, students participated in the on-going discussion of scientific and administrative issues, helping to enhance their understanding of the function and structure of the U-M CEHC while also providing them with opportunities to obtain feedback from junior and senior faculty on their own projects.

Future Activities:

As data and biological specimens become available, we will work with the P30 NIEHS Core Center at the University of Michigan to make data and biological specimens available through a searchable web-based kiosk. Additionally, through the Michigan Regional Comprehensive Metabolomics Research Core (MRC)2, metabolomics data from the two human cohorts will also be made available to the public after publication of data, in accordance with NIH guidelines.
