In laboratory studies, the soil isolate Pseudomonas aeruginosa CHL004 (Vesper et al 1996) has been found to concentrated Pb2+ in the cytoplasm by formation of particles that contain Pb2+ and phosphorus. Upon examination of the washed lyophilized cells grown in the presence of lead using x-ray diffraction we have determined that the product formed is pyromorphite. Other experiments suggest that phosphate concentration has an effect on the uptake of lead into the cell. This alludes to the fact there must be a mechanism by which lead is actively transported into the cell. The specific aims of our current research are to understand the mechanism of lead transport and to optimize conditions for uptake in CHL004. The long-term goal of this project is to utilize this bacterium in a field study to determine the plausibility of utilizing Pb2+ uptake and sequestration to remove Pb2+ from Pb2+-contaminated soils. The experimental included the use of different sodium or hydrogen pump inhibitors, which are possibly involved in the heavy metal active transport system (Rensing et al 1998). A control bacterium was utilized for all the experiments, namely Pseudomonas aeruginosa strain ATCC 27853, which was be was used for comparison. The results of our research efforts to date will be presented.
