REGIONAL METHODS INITIATIVE FOR ALKYLPHENOL ENDOCRINE DISRUPTERS

Impact/Purpose:

There are four objectives:

Develop use of Solid Phase Extraction for rapid extraction of these compounds form water.

Purify commercially available standards for Nonylphenol and its one and two unit ethoxylates and carboxylates and confirm by Mass Spectroscopy for biological studies.

Develop a qualitative and quantitative LC/MS method for the carboxylates using pure standards.

Develop GC/MS and LC/MS methods for these compounds in fish tissue.

Any method developed within the Central Regional Laboratory will be used to generate data for monitoring investigations of certain waterway systems in Region 5 by our Water Division. They will be responsible for using the data to determine distribution of these compounds in the environment.

Description:

Nonylphenol and the ethoxylates are amenable to GC/MS analysis but the carboxylates are better analyzed using LC/MS. The Central Regional Laboratory (CRL) intends to do this with standards of known purity using a triple quadruple instrument. The purified standards will be used to establish sound acute toxicity values for fathead minnows, C. Daphnia and Hyallela. The existing GC/MS method developed in CRL will be used along with the LC/MS method to analyze fish tissue for the presence of these compounds. This will provide information about their uptake in fish from various waterways in Region 5.
