Description:

This report presents a model developed to study growth and differentiation of primary cultures of ureteric epithelial cells from embryonic C57BL/6N mouse urinary tracts. Single cells were resuspended in medium and plated onto transwells coated with collagen IV and laminin. Basal keratinocyte growth medium-2 with low calcium was supplemented with fetal bovine serum, trace elements, a complex mixture of lipids, a defined mixture of purified hormones and growth factors empirically selected to promote growth and survival of the epithelial cells. These cultures were viable for 23 to 30 days. This is the first successful, long-term in vitro model for primary single-cell embryonic ureteric epithelium and could be used to test chemicals that either slow epithelial proliferation or induce programmed cell death. Modifications to the supplementation of the medium provided a less stringent selection for epithelial versus stromal cells and was also less stimulatory for epithelial cell proliferation. These culture models will allow the study of the role of specific regulators of development and agents that either inhibit or stimulate ureteric epithelial proliferation and/or differentiation.
