ABSTRACT
A measure of sperm chromatin integrity was added to the routine semen end points evaluated in the Teplice Program male reproductive health studies. To address the hypothesis that exposure to periods of elevated air pollution may be associated with abnormalities in sperm chromatin, semen samples were evaluated using the Sperm Chromatin Structure Assay (SCSA). Exposure to periods of high air pollution (exceeding US and Czech air pollution standards) was associated with a significant increase in the percentage of sperm with abnormal chromatin. Ongoing studies are comparing these SCSA results with those obtaining using two additional tests for DNA strand breaks in sperm, the COMET test and the TUNEL test. Together, these tests will provide information about potential risks of paternally-mediated abnormal pregnancy outcome.
